摘  要:目的研究转化生长因子β受体Ⅰ(TβRⅠ)蛋白在红藻氨酸(KA)致癫痫大鼠额叶及海马组织的表达,以探讨TβRⅠ与癫痫的关系。方法雄性SD大鼠随机分成对照组(10只)和模型组(50只),模型组再分为致痫后6h、12h、24h、72h、1w共5个亚组;采用红藻氨酸侧脑室注射建立颞叶癫痫动物模型,用Western Bloting技术在不同时间点检测致痫大鼠额叶及海马组织中TβRⅠ蛋白的表达情况。结果模型组大鼠致痫后额叶及海马组织中TβRⅠ蛋白表达较对照组明显增高,于72h达高峰,1w后逐渐降低;海马组织中TβRⅠ蛋白表达比额叶更明显(P<0.05)。结论 TβRⅠ蛋白在致痫后的额叶和海马组织中大量表达,表明TβRⅠ参与了癫痫早期的发病机制。Objective To study the expression of transforming growth factor-β receptorⅠ(TβRⅠ) protein in frontal lobe and hippocampus tissues of kainic acid(KA)-induced epileptic rats,and then to probe the relationship of TβRⅠ and epilepsy.Methods Male SD rats were randomly divided into control group(n=10) and model group(n=50),which were divided into 5 subgroups(6h,12h,24h,72h and 1w after seizures).Animal models of temporal epilepsy were established in rats by intracerebroventricular injection of KA.Expressions of TβRⅠ protein in frontal lobe and hippocampus tissues within various phases after seizures were observed by Western bloting technology.Results Compared with control group,expressions of TβRⅠ protein both in frontal lobe and hippocampus tissues were significantly increased and reached the peak 3d after seizures,and declined gradually 1w after seizures.Expressions of TβRⅠ protein in hippocampus tissues were more significant than those in frontal lobe(P<0.05).Conclusion KA-induced epileptic rats express abundant TβRⅠ protein both in frontal lobe and hippocampus tissues,indicating that TβRⅠ participates in the pristine nosogenesis of epilepsy.
