摘 要:目的探讨PTEN在急性髓性白血病(AML)中的表达情况及其对预后的影响。方法应用免疫组化(S-P)技术分别检测45例初治AML及40例对照(正常骨髓10例及非恶性血液病30例)骨髓单个核细胞中PTEN的表达,分析PTEN与外周血白细胞数及特异性融合基因的关系,追踪疾病的转归。结果 PTEN基因阳性表达在对照组中非恶性血液病85.35%,与正常人90%比较,差异无显著性(P>0.05)。45例初治AML的PTEN表达阳性率为57.17%,明显低于对照组,差异有显著性(P<0.01)。PTEN的低表达与外周血白细胞计数及骨髓中白血病细胞的比率呈正相关(P<0.05);与患者的年龄、性别及与染色体预后分组无明显相关性(P>0.05)。结论 AML患者存在PTEN基因表达异常,PTEN可能也是影响AML患者预后的生物学指标之一。Objective To study the expression of PTEN in acute myeloid leukemia(AML) and its effect on the prognosis.Methods Immunohistochemistry(S-P) was used to detect PTEN expression in bone marrow mononuclear cells of untreated AML(45 cases) and controls(40 cases,including 10 cases of normal bone marrow and 30 cases of non-hematologic malignancies).Analysis was conducted on the association of PTEN with the amountly of peripheral blood leukocytes and specific fusion gene and prognosis of the disease was traced.Results PTEN was positive expressed in 85.35% of the hematologic malignancies patients and 90% of the normal,which showed no significant difference(P0.05).The rate of PTEN expression in 45 cases of newly diagnosed AML was 57.17%,which was much lower than the control group(P0.01).Low expression of PTEN was correlated positively with the amount of peripheral blood leukocytes and the ratio of leukemic cells in bone marrow(P0.05),but bore no relation to such parameters as age,sex and prognostic groups of chromosome(P0.05).Conclusion PTEN gene is abnormally expressed in AML patients,and it could be a valuable marker for the prognosis of AML patients.
