摘 要:目的建立短梗五加果提取物中总黄酮与总酚及原儿茶酸、绿原酸、金丝桃苷3种指标成分的含量测定方法。方法采用紫外-可见分光光度法:以芦丁为对照品,AlCl3显色,在273 nm处测定短梗五加果提取物总黄酮含量;以绿原酸为对照品,Al(NO3)3显色,在520 nm处测定短梗五加果提取物总酚含量。采用HPLC法测定3种指标成分含量:色谱柱为Hypersil ODS2柱(150 mm×4.6 mm,5μm),流动相为甲醇-乙腈-水-冰醋酸溶液,梯度洗脱,流速为1.0 mL.min-1,柱温40℃,检测波长254 nm。结果紫外-可见分光光度法:芦丁质量浓度在5.00~40.00 mg.L-1(r=0.9998)内、绿原酸质量浓度在4.98~59.70 mg.L-1(r=0.999 9)内线性关系良好,芦丁、绿原酸的平均回收率分别为101.8%1、00.7%,RSD分别为1.1%、0.74%。HPLC法:3种指标成分线性范围分别为:原儿茶酸0.1~2.5 mg.L-1、绿原酸16.8~420 mg.L-1、金丝桃苷3.68~92 mg.L-1r,均为0.999 9,平均回收率为96.8%~101.2%,RSD分别为1.7%、1.2%和0.99%。结论两方法简便、准确、重复性好,可综合用于短梗五加果提取物的质量控制。Objective To establish methods for the determination of total flavonoids and total phenols and an HPLC method for simultaneous determination of protocatechuic acid, chlorogenic acid and hyperoside in the extract of fruits of Acanthopanax sessiliflorus. Methods UV-Vis spectrophotometry methods were used to determine the contents of the total flavonoids and total phenols. The absorbance was measured at the wavelength of 273 nm and 520 nm using rutin and chlorogenic acid as standards, AlCl3 and Al ( NO3 ) 3 as chromogenic agents,respectively. The HPLC analysis was carded out on a Hypersil ODS2 (150mm×4.6mm,5μm)column, with a linearity gradient of methanol-acetonitrile-water-acetic acid at the flow rate of 1.0 mL·min^-1. The detective wavelength was set at 254 nm, and the column temperature was set at 40 ℃. Results UV-Vis spetrophotometry was used, the linear range was 5.00- 40. 00 mg·L^-1 ( r = 0. 999 8 ) for rutin,the average recovery was 101.8% with RSD of 1.1%. The linear range was 4. 98-59.70 mg·L^-1( r = 0. 999 9)for chlorogenic acid, the average recovery was 100. 7% with RSD of 0. 74%. The HPLC method was used, and the linear ranges of protocatechuic acid, chlorogenic acid and hyperoside were 0. 1 - 2. 5 mg·L^-1 ,16. 8-420 mg·L^-1 ,3.68-92 mg·L^-1 ,respectively,with r of 0. 999 9. The recoveries ranged from 96. 8% to 101.2% with RSD of 1.7% ,1.2% and 0. 99%. Conclusions The methods are simple,accurate and reproducible. They can be used for quality control of the extract of A. sessiliflorus fruits.
