摘  要:目的研究丹参单体(IH764-3)和三氟拉嗪对体外培养的兔视网膜色素上皮(retinal pigment epithelial,RPE)细胞增殖的抑制作用。方法将传代培养的RPE细胞分别加药培养24h、48h、72h、96h、120h,应用二甲基偶氮唑盐(MTT)比色法检测不同剂量丹参单体和三氟拉嗪对RPE细胞的抑制作用,进一步检测两药半数抑制剂量(IC50)联合应用对RPE细胞的抑制作用。结果丹参单体和三氟拉嗪对RPE细胞的增殖均有抑制作用,呈时间及剂量依赖性,各剂量及各时间点相比差异均有显著统计学意义(均为P<0.01),丹参单体的IC50为15.22μmol·L-1(2.4mg·L-1),三氟拉嗪的IC50为18.09μmol·L-1。联合用药120h对RPE的抑制率达87.23%,联合用药后各时间点与单独应用丹参单体及三氟拉嗪的抑制率比较,差异也均有显著统计学意义(均为P<0.01)。结论丹参单体和三氟拉嗪对RPE细胞的增殖均有抑制作用,两药联合后可产生协同作用,为以后两种药物的联合体内试验提供了一定的实验基础。Objective To investigate the inhibitive effect of IH764-3 and trifluoperazine(TFP) on the proliferation of retinal pigment epithelial(RPE) cells cultured in vitro.Methods Cultured RPE were exposed to different concentrations of IH764-3 and TFP.The inhibitive effects of IH764-3 and TFP on the proliferation of RPE cells of each group were examined at 24 hours,48 hours,72 hours,96 hours and 120 hours after the exposure,and the IC50 of the two drugs were calculated.Results Both IH764-3 and TFP had significant inhibitive effect on the proliferation of RPE cells in both dose-dependent and time-dependent manners,there were significant differences in dose and time point(all P0.01).The IC50 of IH764-3 on RPE cells was 15.22 μmol·L-1(2.4 mg·L-1),while TFP was 18.09 μmol·L-1.At 120 hours after combined administration of the two drugs,the inhibitory rate arrived at 87.23%.The inhibitory effect of combination of two drugs was much better than single use of IH764-3 or TFP,there were significant differences(all P0.01).Conclusion IH764-3 and TFP have an inhibitive effect on the proliferation of RPE cells in vitro,the combined application of two drugs can produce synergetic effect,which prefer the experimental basis for combined application of two drugs in vivo.
