摘  要:目的检测胃癌组织中p27基因的5’CpG岛甲基化状态,并探讨p27基因甲基化状态与胃癌生物学特性的关系。方法利用甲基化特异性PCR测定49例胃癌组织,20例癌旁组织以及lO例正常胃黏膜组织中p27基因的甲基化状态。结果胃癌组p27基因甲基化率为49%,癌旁组p27基因甲基化率为25%,正常胃黏膜组p27基因甲基化率为0。胃癌组p27基因甲基化率明显高于正常胃黏膜组(P〈0.05);淋巴结转移组p27基因甲基化率为61%,显著高于无淋巴转移组的28%(P〈0.05)。p27基因甲基化率与患者年龄、性别、肿瘤组织大小、部位无关(P〉0.05)。结论p27基因甲基化可能导致p27mRNA的失转录,并与胃癌的发生、转移相关。Objective To detect methylation status of the 5'CpG island locating in the promoter region of p27 gene in gastric carcinoma and to analyze the relationship between the aberrant methylaion of p27 gene and the bionomics of gastric carcinoma. Methods Methylation-specific PCR were used to detect the methylation status of p27 in gastric carcinoma( n = 49), adjacent tissue of carcinoma (n = 20)and normal tissue(n = 10), respectively. Results The mathylation rate is 49% (24/49) for gastria cancer, 25% (5/20) for tumor-sideward mucosa and 0 (0/10)for normal mucosa. There was a significant positive correlation between gastric carcinoma specimens and normal mucosa specimens ( P 〈 0.05 ). There was no significant diference between normal gastric group and tumor-sideward mucosa group ( P 〉 0. 05) and no significant diference between tumor-sideward mucosa group and tumor group ( P 〉 0. 05 ). The mathylation rate of p27 gene with lymph node metastases (61% ) are higher than those without metastasis (28 % ) (P 〈 0.05). And there is no relation on patients' age, sex, tumor size and location. Conclusion p27 gene methylation of the p27 gene is correlated to oncogenesis and metastasis of gastric carcinoma and could lead to the loss of p27mRNA transcription.
