摘  要:目的探讨双侧杏仁核去5-羟色胺(5-HT)支配后小鼠的行为学变化,投射到杏仁核的5-HT能神经纤维的精确来源以及杏仁核神经元的异常功能活动,为验证杏仁核-中缝核群5-HT在抑郁症发病机制中的联系提供实验资料。方法雄性昆明小鼠32只,随机分为对照组和实验组,每组各16只,应用脑立体定位注射技术向双侧杏仁核注射5,7-双羟色胺(5,7-DHT,5μg/侧),悬尾实验和强迫游泳实验检测抑郁行为;脑石蜡切片H—E染色检测5-HT能投射神经元的位置;脑冰冻切片免疫组织化学染色检测杏仁核FosB/AFosB的表达。结果①行为学检测:实验组小鼠强迫游泳无动时间和悬尾无动时间较对照组显著减少(P〈0.01);②H—E染色:实验组小鼠中缝背核尾侧份与中缝正中核嘴侧份的部分神经元胞体皱缩变形,胞质呈深紫伊红色染色,尼氏小体溶解或消失,胞核缩小或消失;③免疫组织化学染色:FosB/△FosB阳性神经元在实验组小鼠杏仁核明显增多,与对照组相比差异显著(P〈0.01)。结论杏仁核的5-HT能纤维投射主要来自中缝背核的尾侧和中缝正中核的嘴侧;单独的杏仁核去5-HT支配即可引发小鼠产生抑郁样行为;杏仁核内的即早基因FosB/△FosB在抑郁样行为形成的相关精神病理学机制中占有重要地位。Objective To investigate the ethological changes following the 5 - hydroxytryptamine (5 - HT) depletion in bilateral amygdalae in mice, the precise resource of 5 - HT - ergic nerve fibers projecting to the amygdala and the functional abnormalities of amygdala neurons, so as to provide experimental data for the verification of the association of 5 - HT of amygdala - raphe nuclei in the pathogenesis of depression. Methods Thirty - two male Kunming mice were randomized into the control group and the experiment group (n = 16 each). The murine bilateral amygdalac received sterotaxic injection of 5,7 -dihydroxytryptaminc (5,7 -DHT, 5 μg/L each). Forced swimming and tail suspension experiments were applied to detect depressive behavior in mice ; hematoxylin - eosin ( H - E) staining of paraffin sections of the murine brain was performed to detect the sites of 5 - HT - ergic fiber projection; the expression of FosB/AFosB in the amygdala was detected by immunohistoehemistry staining of coronal sections of the mouse brain by a freezing microtome. Results 1. Ethological detection: compared with the control group, the immobility time during forced swimming and tail suspension significantly decreased in the experiment group (P 〈 0.01 ). 2. H -E staining: shrinkage and deformation of the neurons at the caudal part of raphe dorsal nucleus and rostral part of median raphe nucleus, dark purple in cy- toplasm by H - E staining, dissolution or disappearance of Nissl bodies, nuclear shrinkage (karyopycnosis) or disappear- ance; 3. Immunohistochemistry staining: marked increase in the expression of FosB/△FosB -positive neurons in the amygdale in the experiment group ( P 〈 0.01 ) compared to the control group. Conclusion 5 - HT - ergic fiber projec tion in the amygdala mainly originates from the rostrol part of raphe dorsal nuclei and the rostral aspect of median raphe nuclei. 5 - HT depletion in individual amygdala can trigger depression - like behaviors in mice.
