摘 要:目的探讨黄芪三七合剂对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的作用及机制。方法将雄性SD大鼠60只随机分为3组:UUO组(UUO,n=20)、假手术组(SOR,n=20)、黄芪三七合剂组(A&R,n=20)。从术后24小时开始,A&R治疗组每天分别予A&R(3ml/d)灌胃;UUO组和SOR组予以同等体积生理盐水灌胃,连续14天。于术后3,7,14天分别随机处死各组大鼠。行Masson染色观察肾间质病理学改变;化学比色法测定肾组织匀浆中羟脯氨酸的含量;肾脏组织匀浆检测活性氧自由基(OH.)、脂质过氧化产物丙二醛(MDA)、超氧化物歧化酶(SOD)。采用Western蛋白印迹、免疫组化技术从蛋白质水平检测血红蛋白氧化酶(Heme Oxygenase 1,HO-1)的表达。结果 UUO组肾脏呈进行性肾间质纤维化及肾间质胶原沉积的改变(P<0.05)。A&R组与UUO组比较在不同程度上减轻了胶原在肾间质的沉积和肾脏病理损害。与SOR组相比,UUO组大鼠OH.和MDA高于SOR组,SOD低于SOR组。A&R治疗组均不同程度地改善了上述改变。与SOR组相比,HO-1蛋白的表达UUO组3d开始升高,7d仍有较高的表达,14d回落。A&R治疗组对上述改变作用不明显。结论 A&R对UUO所致的大鼠肾脏纤维化的形成有一定的改善作用,并有一定的抗氧化应激的作用。但对HO-1的作用不明显。Objective The present study was undertaken to investigate the effects of astragalus and radix notoginseng(AR) on renal fibrosis in rats with unilateral ureteral obstruction(UUO).Methods The rats were randomly divided into 3 groups: sham operation(n=20),UUO(n=20) and UUO with(AR) treatment(n=20).The rats were sacrificed at d 3,7,14 respectively,after the surgery.The histological changes of renal interstitial tissues were observed by Masson staining.Total kidney collagen content was determined by measuring the amount of hydroxyproline.Hydroxy radical(OH·.),ma1ondia1dehvde(MDA),and superoxide dinmutase(SOD) contents were determined by biochemical methods.The Protein of Heme Oxygenase 1(HO-1) expression was assessed by immunohistochemistry and western blot analysis.Results The results demonstrated that AR significantly inhibited interstitial fibrosis including tubular injury as well as collagen deposition(P0.05).Compared with the UUO group,the levels of OH·.and MDA was decreased obviously in the AR treatment group(P0.05).Inversely the level of SOD was obviously increased(P0.05).The expression of HO-1 in tubular and interstitial cells was increased in UUO control kidneys at d 3 and was maintained at d 7,then declined at d 14(P0.05 vs.SOR).However,AR on the role of HO-1 is not obvious.Conclusion AR can inhibit renal interstitial fibrosis in rats with UUO,possibly via an antioxidant mechanism.
