塞来昔布和洛伐他汀对鼻咽癌细胞Caveolin-1的表达及下游信号通路的影响

摘　　要：目的探讨塞来昔布和洛伐他汀对鼻咽癌细胞Caveolin-1表达及下游信号分子的影响。方法体外培养鼻咽癌细胞CNE-2,经塞来昔布和洛伐他汀处理后,MTT检测其增殖情况。随后用IC50浓度的塞来昔布和洛伐他汀与CNE-2细胞孵育,Western blot检测Caveolin-1的表达以及Akt、ERK1/2和STAT3的磷酸化情况。结果 CNE-2细胞经洛伐他汀和塞来昔布处理24h后,IC50值分别为20μM和40μM;塞来昔布或洛伐他汀单独处理CNE-2细胞24h,能轻微下调Caveolin-1的表达,当两者联合使用时,能使Caveolin-1水平进一步降低,24h后几乎完全抑制Caveolin-1的表达;洛伐他汀、塞来昔布能抑制血管内皮生长因子诱导的Akt、ERK1/2和STAT3磷酸化,当两者共同处理CNE-2细胞时,能进一步抑制其磷酸化水平。结论塞来昔布能协同洛伐他汀下调鼻咽癌细胞Caveolin-1的表达,从而可能影响下游信号分子的激活。Objective To explore the effect of celecoxib and lovastatin on the expression of nasopharyngeal carcinoma cells caveolin-1 and activation of the downstream signaling molecules.Methods Nasopharyngeal carcinoma cell line CNE-2 cells were cultured in vitro,MTT test was used to detect the IC50 of celecoxib and lovastatin on cell proliferation,then cells were incubated with IC50 concentrations of celecoxib and lovastatin,and Western blot was used to detect the expression of caveolin-1 and the phosphorylation levels of Akt,ERK1 / 2 and STAT3.Results CNE-2 cells after lovastatin and celecoxib 24h treatment,the IC50 values were 20μM and 40μM respectively.PC-3 cells treated with lovastatin or celecoxib separately 24h,slight down-regulated caveolin-1 expression.When the two drugs were jointly used,they could further reduce the expression level of caveolin-1.However,after 48h of coincubation,the caveolin-1 was hardly expressed.Lovastatin and celecoxib could also inhibited EGF-induced Akt,ERK1/2 and STAT3 phosphorylation.When the two work together on CNE-2 cells,they could further inhibit their phosphorylation.Conclusions Celecoxib and lovastatin synergistically down-regulate nasopharyngeal carcinoma cells the expression of caveolin-1,and then may affect the activation of downstream signaling molecules.
