β-tubulinⅢ及survivin蛋白在胃癌中的表达及相互关系的研究

摘　　要：目的 探讨β-微管蛋白Ⅲ（β-tubulinⅢ）、生存素（survivin）在各期胃癌组织中的表达及两者之间的相互关系.方法 检测60例不同分期、分化程度胃癌组织标本中（胃癌组） β-tubulinⅢ、survivin的表达情况,并与30例正常无病变的胃黏膜标本（对照组）作比较.利用Bradford法定量检测β-tubulinⅢ与survivin在不同分化程度胃癌组织中的表达.结果 胃癌组β-tubulinⅢ阳性表达率36.7%,明显高于对照组的0%（P＜0.01）;胃癌组survivin阳性表达率58.3%,明显高于对照组的3.3%（P＜0.01）.β-tubulinⅢ在高、中、低分化胃癌组织中的表达率分别为11.11%、33.33%、44.44%,而survivin在高、中、低分化胃癌组织中的表达率分别为33.33%、53.33%、66.67%,差异有统计学意义（P＜0.05）.胃癌组织中β-tubulinⅢ与survivin表达的相关性,经Spearman分析两者呈正相关（P＜0.05）.结论 胃癌组织标本中存在β-tubulinⅢ、survivin的高水平表达,且与组织分化程度有关,两者表达呈正相关,β-tubulinⅢ、survivin可能共同导致了胃癌肿瘤细胞的异常增殖和异常凋亡,β-tubulinⅢ和survivin抑制剂与基因治疗可能会成为今后胃癌防治的一个主要方向.Objective To investigate the expression of β -tubulin III and survivin in gastric carcinoma and the correlation between them. Methods Expression of β-tubulin III and survivin was detected with immuno-histochemistry in 60 tissue specimens of gastric cancer and 30 specimens of normal gastric mucosa （control）, Results The positive rates of β -tubulin III and survivin in gastric cancer were 36.7% and 58.3% respectively, which were significantly higher than those in normal gastric mucosa （0% and 3.3% respectively, P 〈 0.01）. Spearman analysis showed that expression of β -tubulin Ill was positively correlated with survivin in gastric cancer tissues （P〈 0.05）. Conclusion Gastric cancer has a high expression level of β -tubulin III and survivin, which might be involved in abnormal proliferation and apoptosis of gastric tumor cells.
