摘 要:目的采用可生物降解的壳聚糖、海藻酸钠和胶原蛋白制备一种复合微粒并评价其安全性能。方法采用乳化交联技术混合以上几种材料制成一种结构疏松的复合微粒;粒径分析仪测定其粒径;扫描电镜观察其超微结构;在模拟体液中测定其吸水率;MTT法观察其细胞毒性,腹腔注射观察其急性毒性;体外降解实验考察其生物降解性能。结果该复合微粒呈白色粉体状,结构疏松,形态圆整,密度为(140±14)mg/cm3,粒径分布为2~40μm;复合微粒在模拟体液中迅速溶胀,逐渐降解,2 min时溶胀率达到2293%,4周后的降解率为40%;实验初期,微粒对细胞的增殖和小鼠的行动均有一定的抑制作用,并随时间的延长而减弱或消失。结论制备的复合微粒粒径分布窄,吸水性好,可生物降解且具有较好的安全性,可望作为一种新型的粉体生物敷料。Objective To prepare a new kind of degradable composite powder(CP) based on chitosan,sodium alginate and collagen protein,and to evaluate its safety.Methods By the technology of emulsification and cross-linking the CP was prepared with chitosan,alginate and collagen protein as main materials.The size distribution of the CP was determined by powderdiameter analysator,and the surface of CP was observed under SEM.The swelling characteristics and the degradation in vitro of CP were investigated in the simulative body fluid.The cytotoxicity of the CP for L929 fibroblasts was monitored using MTT assay and the acute toxicity for mice was tested by peritoneal injection of the extract of CP.Results The CP prepared had a loose structure with a density of(140±14) mg/cm3 and size distribution of 2~40 μm.The powder quickly swelled and gradually degraded in the simulated body fluid,with a water uptake ratio of 2293% in 2 min and a degradation ratio of 40% after 4 weeks.The results of MTT assay and acute toxicity test revealed that the CP displayed slight inhibition to the cells proliferation and the action of mice at initial stage,but the inhibition would vanish as time went on.Conclusion The CP made of chitosan,alginate and collagen protein possesses confined size distribution,good water uptake ability,biodegradability and bio-safety,so it might be one of promising and safe biomaterial with potential applications.
