摘 要:目的观察SnoN蛋白在大鼠肝纤维化进展过程中的动态表达变化及其意义。方法 SD大鼠分两组:正常对照组6只,模型组24只,二甲基亚硝胺(DMN)腹腔注射制备大鼠肝纤维化模型。造模后分别在第2周、4周、6周、8周取大鼠血及肝脏标本,HE和Masson染色并光镜下观察各组大鼠肝组织病理变化;RT-PCR检测SnoN、Smurf 2、TGF-β1及Ⅰ型胶原纤维(COL-1)在造模过程中的动态变化。结果模型组第4周、6周肝纤维化明显;模型组血清ALT、AST升高,第4周时达高峰;Alb逐步下降,6周时最低,各时间点与对照组比较均有统计学差异(P<0.005)。造模后,SnoN表达进行性降低,第8周达到最低,Smurf 2表达量逐渐增加,在第6周达最高峰,且明显高于对照组,各模型组与对照组相比均具有显著差别(P<0.05或0.01);TGF-β1表达水平逐步增高,于第4周时达高峰,各模型组与对照组相比均具有统计学差异(P<0.01)。结论 SnoN蛋白表达的降低可能是肝纤维化中的一个重要环节,值得进一步深入研究。Objective To observe the changes of SnoN expression in liver tissues of hepatic fibrosis rats,and to elucidate the role ofSnoN protein in the hepatic fibrosis. Methods Hepatic fibrosis was induced by intraperitoneal of DMN in SD rats. The model animals were divided into 2 groups:the negative control group and the control group. After the animal model was made,blood sample and liver tis- sue at 2,4,6 and 8 weeks were collected. The expressions of SnoN,Smurf 2 mRNA,TGF - β1 ,COL - 1 were detected by RT - PCR. ALT, AST, Alb, Tbil were detected by biochemistry methods. The liver morphology was checked on HE and Masson sections under light microsco- py. Results Alb,AST,ALT were raised obviously in all model groups compared to the normal groups. SnoN protain was decreased in the 4 weeks,6 weeks,and the 8 weeks groups. TGF - β1 ,Smurf 2 had a contradictory result(P 〈 0.01 ). Conclusion With the development of hepatic fibrosis,the SnoN protain decreases gradually. The expression of SnoN protain may play a important role in hepatic fibrosis,and it should be researched in- depth.
