系统性红斑狼疮患者血液中PRDM1和CD138＋浆细胞表达的研究

摘　　要：目的;探讨系统性红斑狼疮(SlE)患者和正常人之间PRDM1和CD138+浆细胞的差异。方法:40例SLE患者血液标本,30例健康体检血液标本,抽取RNA、利用逆转录聚合酶链反应(RT-PCR)技术研究PRDMl基因表达;收集外周血单个核细胞,利用流式细胞技术研究CDl38’浆细胞量的差异。结果:SLE患者组血液中的PRDM1表达量明显高于健康体检组(p<0.05),并且CD138+浆细胞的量也多于健康体检组。结论:SLE实验组中的PRDM1明显高于体检对照组,CD138+浆细胞的量也高于对照组,提示SLE患者中浆细胞数量的增多与PRDMl高表达相关,这为进一步了解SLE发病机制提供了一个新的方向和靶点。Objective： To determine the difference of PRDM - 1 and CD138 ＋ plasma cells in between patients with systemic lupus erythematosus （SEE） and normal. Methods： forty cases of SLE patient blood samples, thirty cases of healthy blood samples, extract RNA, using reverse transcription polymerase chain reaction （RT- PCR） technique to study PRDMI gene expression; collection of peripheral blood mononuclear cells, differences in the amount of CD138＋ plasma ceils by flow cytometry. Result： The PRDM1 expression in SLE patients blood was significantly higher than that healthy group （p 〈 0.05）, and the amout of CD138＋ plasma cells are also more than healthy group. Conclusion： PRDMI and the amount of CD138＋ plasma cells in SLE study group is significantly higher than control group, indicating that the high PRDM1 expression and the number of plasma cells are relevant, this will provides a new direction and target to study SLE.
