摘 要:从土壤中筛选产阿拉伯胶酶的微生物菌株,并通过紫外诱变选育后得到高产突变菌株ZHB05F,依据菌落和孢子形态特征初步鉴定为镰刀(Fusarium sp.)。通过单因素试验,优化了产酶培养基的主组分的浓度和pH值,得到最佳的产酶发酵培养基组成为:阿拉伯胶30 g/L,(NH_4)_2SO_4 8g/L,K_2HPO_4 1g/L,MgSO_4·7H_2O 0.5 g/L,KCl 0.5g/L,FeSO_4·7H_2O 0.01g/L,H_3BO_3 0.01 g/L,pH 5~6。菌株ZHB05F在优化培养基组成下进行产酶发酵,酶活达到147.7 U/mL。A fungi strain, which could produce arabic gum degrading enzyme, was screened out from soil, and then was identified as Fusarium sp. by the characteristic of its morphology of clone and spore. After mutagenesis by ultraviolet, a high-yielding strain designated ZHB05F was selected out. Further, the composition of fermentation medium for enzyme production was optimized. The results showed that the optimal fermentation medium was consisted of arabic gum 30g/L,(NH):SO4 8g/L,K2HPO4 1g/L,MgSO4·7H2O0.5g/L,KCl0.5g/L,FeSO4·7H2O0.01g/L,H3BO3 0.01g/L,pH5~6 with pH 5--6, the activity of enzyme was up to 147.7U/mL produced by the strain ZHB05F.
