摘 要:目的探讨TDGF-1基因小干扰RNA(small interfering RNA,siRNA)对人鼻咽癌细胞迁移侵袭的影响。方法根据TDGF-1基因序列特点设计了siRNA,转染人鼻咽癌CNE-2细胞,分别采用实时定量PCR和蛋白质印迹法检测TDGF-1基因的mRNA和蛋白水平,以软琼脂集落形成试验观察癌细胞锚着不依赖性增殖能力,以划痕试验方法评测癌细胞的迁移能力;用Boyden模型观察癌细胞侵袭能力。结果 TDGF-1基因siRNA转染组TDGF-1 mRNA和蛋白水平明显下调,且呈浓度和时间依赖性(均P<0.001)。体外试验发现,TDGF-1 siRNA转染可有效抑制鼻咽癌细胞集落生长、侵袭和迁移能力,且与浓度相关(均P<0.005)。结论 TDGF-1在鼻咽癌细胞生长、迁移、侵袭中起着重要的作用;采用TDGF-1 siRNA转染可抑制鼻咽癌细胞侵袭。Objective To study the effects of TDGF-1 small interfering RNA (siRNA) on invasion and migration of human nasopharyngeal cancer cells. Methods After human nasopharyngeal cancer cell line CNE-2 was transfected by TDGF-1 siRNA, the mRNA and protein expression level of TDGF-1 were determined by real time-PCR and Western blot assay, respectively. The anchorage-independent growth was examined by clone formation in soft agar, and the ability of migration and invasion was evaluated by wound-healing assay and boyden chamber model, respectively. Results The siRNA could down-regulate the level of mRNA and protein of TDGF-1 in a dose- and time- dependent manner. Suppression of TDGF-1 expression could inhibit anchorage-in- dependent growth in a dose-dependent manner, and invasion, and migration ability of nasopharyngeal cancer cell. Conclusions TDGF-1 gene might play an important role in the development of human nasopharyngeal cancer. Down-regulated TDGF-1 by siRNA could inhibit growth, invasion and migration of human nasopharyngeal cancer cells.
