摘 要:目的观察吗啡条件性位置偏爱(CPP)大鼠消化系统食管、胃、十二指肠P2Y2受体mRNA表达的变化,探讨阿片类精神依赖下消化系统异常的分子机制。方法吗啡剂量递增法颈背部皮下注射建立大鼠吗啡CPP模型(模型组),生理盐水对照组则予注射生理盐水,其余处理同模型组。实时荧光定量PCR分别检测大鼠食管、胃、十二指肠P2Y2受体mRNA的表达水平,采用2-△△Ct方法进行比较分析。结果与生理盐水对照组比较,模型组大鼠在白箱的停留时间明显延长(P<0.01);食管、胃、十二指肠P2Y2受体mRNA表达水平下调,分别仅为生理盐水对照组的0.41、0.33和0.14倍。结论 P2Y2受体mRNA表达下调可能与吗啡精神依赖过程胃肠道功能的失调有关。Objective To examine the mRNA level of P2Y2 receptor in the esophagus,stomach and duodenum in rat model of conditioned place preference(CPP),and to investigate the molecular mechanism of dysfunction of digestive system of rats in opioid-psychic dependence.Methods The rat model of CPP was constructed by subcutaneous injection of morphine into neck and back by dose escalation.The real-time quantitative RT-PCR was applied to assess the expression of P2Y2 receptor mRNA in esophagus,stomach and duodenum.The result was analyzed by the 2-△△Ct test.Results Compared to the saline control rats,significantly prolonged detention time in white compartment for the model group was notably observed(P0.01).Moreover,the expression levels of P2Y2 receptor mRNA in esophagus,stomach,duodenum of model rats were 0.41,0.33 and 0.14 fold,respectively,of the NS control group.Conclusion Lower expression of P2Y2 mRNA may correlate with gastrointestinal function disorder in morphine-psychic dependent rats.
