摘 要:目的检测不同发病程度的癫痫患者颞叶或海马中脑源性神经营养因子(BDNF)及其酪氨酸激酶B受体(TrkB)差异性表达情况,探讨其在癫痫发病机制中的作用。方法取82例癫痫患者颞叶或海马脑组织,用免疫组化方法检测BDNF及其受体TrkB的表达,分析不同病程和发作频率患者脑组织中BDNF及TrkB的表达差异。结果癫痫患者与非癫痫患者比较,颞叶或海马中BDNF及其受体TrkB的含量明显增加。随着患者病程的延长和发作频率的增高,BDNF及其受体TrkB的含量逐渐增加。结论癫痫患者脑组织中BDNF和Trkb表达增高,可能是促进癫痫反复发作的原因之一。Objective To investigate the expression of brain derived neurotrophic factor(BDNF) and its receptor TrkB in the brain tissues of epilepsy.To discuss its function in the epilepsy pathogenesis.Methods 82 cases of hippocampus and temporal lobe brain tissues were studied.The expression of BDNF and TrkB were investigated by Immunohistochemistry.The patients with different pathogenesis and seizure frequency were analyzed.Results The expression of BDNF/TrkB protein was significantly increased in temporal lobe cortex and hippocampus of intractable epilepsy respectively as compared with controls in the same regions.The expression of BDNF/TrkB protein was gradually increased along with extension of pathogenesis and periodicity in temporal lobe cortex and hippocampus of epilepsy.Conclusion It is indicate that the overexpression of BDNF and TrkB in the brain tissues of patients with intractable epilepsy might play an important role in the generation and development of epilepsy.
