摘 要:目的研究丹参单体(IH764-3)对体外培养的兔视网膜色素上皮(retinal pigment epithelium,RPE)细胞增殖的抑制作用。方法将传代培养的RPE细胞分别加入含丹参单体1、2、3、4、5 mg/L培养液培养24、48、72、96、120h,应用噻唑蓝比色法检测不同浓度丹参单体对RPE细胞的抑制作用。结果丹参单体对RPE细胞的增殖有明显抑制作用,24、48h时4mg/L组与5mg/L组抑制率相比差异无统计学意义(P>0.05),其余时间点时各组之间抑制率相比差异有统计学意义(P均<0.01),5mg/L组72h与96h及96h与120h时抑制率差异无统计学意义(P>0.05),其余各组各时间点之间抑制率相比差异有统计学意义(P均<0.01),丹参单体120h时的IC_(50)剂量为2.4mg/L。结论丹参单体对RPE细胞的增殖有抑制作用,呈时间依赖性及剂量依赖性,可能是一种有价值的防治增殖性玻璃体视网膜病变的药物。Objective To investigate the inhibitory effect of Salvia miltiorrhiza monomer( IH764 - 3) on the proliferation of retinal pigment epithelium (RPE) cells in vitro. Methods Cultured RPE cells were exposed to different concentrations of IH764 -3 namely 1,2,3,4 and 5 mg/L. The inhibitory effect on RPE cells by different concentrations of IHT64 -3 was tested with methyl thiazolyl tetrazolium colorimetric (MTT) mearuments 24,48,72,96 and 120 hours after exposure. Results IH764 -3 had a significant inhibitory effect on the proliferartion of RPE in vitro. There were no statistical significances in terms of inhibitory rate between 4mg,/L group and 5 mg/L group at either 24 or 48 hours ( P 〉 0.01 ). There were statistical significances among different concentration groups in any other time (with P 〈 0.01 in all comparisions). There were statistical significances in terms of inhibitory rate neither in comparision of 72 and 96 hours nor in that of 96 and 120 hours in 5 mg/L group, while there were significant differences among each other measuring time points within each individual group. The ICs0 dose of IH764 -3 was 2.4mg/L. Conclusion IH764 -3 has an inhibitory effect on the proliferation of RPE cells in vitro in a time - dependent as well as concentration - dependent manner, indicating that IH764 - 3 is possibly a valuable instrument in the treatment as well as prevention of proliferative vitreoretinopathy (PVR).
