摘 要:目的观察干扰素α-2b(IFNα-2b)对人眼Tenon囊成纤维细胞纤维化作用的影响。方法取青光眼患者滤过术中剪下的Tenon囊组织,在体外进行成纤维细胞原代及传代培养。采用半定量反转录聚合酶链反应(RT-PCR),研究0、125、250、500和1 000 IU/mL浓度的IFNα-2b对体外培养的成纤维细胞转化生长因子β(TGF-β1、TGF-β2)、I型胶原蛋白、纤维连接蛋白表达的影响,以及IFNα-2b对TGF-β2促纤维化作用的影响。以人甘油醛-3-磷酸脱氢酶(GAPDH)作为内参照,计算产物/GAPDH像素值。结果与对照组(TGF-β1:0.50±0.03;TGF-β2:0.53±0.10)相比,250~1 000 IU/mL浓度IFNα-2b组均能明显抑制人Tenon囊成纤维细胞TGF-β1(0.45±0.18、0.45±0.13、0.37±0.11,P<0.05、0.05、0.01)和TGF-β2(0.47±0.12、0.48±0.21、0.42±0.14,P<0.01、0.05、0.01)表达。与对照组(I型胶原蛋白:0.68±0.08;纤维连接蛋白:0.72±0.09)相比,125~1 000 IU/mL浓度IFNα-2b组均能明显抑制I型胶原蛋白(0.53±0.17、0.54±0.20、0.54±0.18、0.44±0.06,P<0.01)和纤维连接蛋白(0.68±0.14、0.67±0.19、0.64±0.07、0.58±0.11,P<0.01)表达。在10μg/L TGF-β2刺激下,与对照组(Ⅰ型胶原蛋白:1.00±0.06;纤维连接蛋白:0.85±0.16)相比,1 000 IU/mL组IFNα-2b能够显著抑制I型胶原蛋白(0.78±0.09,P<0.01)表达;125~1 000 IU/mL组IFNα-2b均能显著抑制纤维连接蛋白(0.74±0.22、0.68±0.18、0.68±0.15、0.63±0.10,P<0.01)表达。结论 IFNα-2b能够通过减少TGF-β1、TGF-β2的表达,减少Ⅰ型胶原蛋白及纤维连接蛋白的表达;还能够拮抗外源性TGF-β2引起的Ⅰ型胶原蛋白及纤维连接蛋白表达升高。Objective To investigate the effect of interferon α-2b (IFNα-2b) in the fibrosis of human Tenon's fibroblasts. Methods Fibroblasts were cultured from Tenon's tissue of a glaucoma patient after trabeeulectomy. The effects of IFNα-2b on the expression of transforming growth factor β1 ( TGF-β1 ), TGF-β2, collagen type Ⅰ and fibronectin mRNA were determined by reverse transeriptase-polymerase chain reaction (RT-PCR) analysis. The effects of IFNα-2b on collagen type Ⅰ and fibroneetin mRNA with the stimulation of TGF-β2 ( 10 μg/L) were also studied. IFNα-2b was added to the culture medium at final concentrations of 0, 125,250, 500 and 1 000 IU/mL respectively. Results The expressions of TGF-β1, TGF-β2 mRNA significantly decreased after the addition of 250 - 1 000 IU/mL IFNα-2b. The expressions of collagen type Ⅰ and fibronectin also significantly decreased at the 125 - 1 000 IU/mL IFNα-2b. The increasing expressions of collagen type Ⅰ and fibronectin mRNA stimulated by 10 μg/L TGF-β2 could be weakened by 1 000 IU/mL and 125 to 1000 IU/mL IFNα-2b respectively. Conclusions IFNα-2b might inhibit the expression of collagen type I and fibronectin mRNA, through decreasing the expression of TGF-β1 and TGF-β2 mRNA. In addition, IFNα-2b could also weaken the increasing expressions of collagen type Ⅰ and fibronectin mRNA stimulated by 10 μg/L TGF-132.
