摘  要:目的:探讨β-catenin表达下调对Tca8113细胞增殖、周期、凋亡和迁移的影响。方法:用脂质体2000将β-catenin siRNA转染舌鳞状细胞癌Tca8113细胞,Western blotting技术检测转染后Tca8113细胞中β-catenin蛋白的表达,CCK-8试剂分析转染后对细胞增殖能力的影响;流式细胞术检测下调β-catenin表达对Tca8113细胞周期及细胞凋亡的影响;Boyden chamber实验分析β-catenin表达下调对Tca8113细胞迁移能力的影响。结果:β-catenin siRNA能明显下调Tca8113细胞中β-catenin蛋白的表达,显著抑制Tca8113细胞的增殖。β-catenin siRNA组中的G0/G1期的细胞百分比明显高于未处理组和对照siRNA组。此外,β-catenin siRNA组中细胞凋亡的比率明显高于未处理组和对照siRNA组,其凋亡变化与caspase-3和bax蛋白表达的上调和bcl-2表达的下降密切相关。与未处理组和对照siRNA组相比,β-catenin siRNA组中Tca8113细胞的迁移能力显著下降,组间具有统计学意义。结论:β-catenin在舌鳞状细胞癌的发生发展中可能具有重要的作用。Objective: To investigate the effect of down-regulation of β-catenin expression on cell proliferation,cell cycle,cell apoptosis and cell migration in Tca8113 cells.Methods: β-catenin siRNA was transfected into Tca8113 cells,and Tca8113 cells were divided into three groups: untreated group,control siRNA group and β-catenin siRNA group.They were harvested 48h after transfected with β-catenin siRNA.Cell proliferation was studied using CCK-8 kit.Effects of downregulation of β-catenin on cell cycle and cell apoptosis were investigated by flow cytometry.Effect of downregulation of β-catenin expression on cell migration was studied using Boyden chamber experiment.Results: β-catenin siRNA obviouslu decreased β-catenin protein expression and significantly inhibited cell proliferation.The percentage of G0/G1 phase in β-catenin siRNA group was significantly higher than that in untreated group or control siRNA group.The percentage of cell apoptosis in β-catenin siRNA group was markedly higher than that in untreated group or control siRNA group.The changes of cell apoptosis were tightly associated with increase of caspase-3 and bax proteins and decrease of bcl-2 protein.Cell number migrated into membrane in β-catenin siRNA group was evidently lower than that in untreated group and empty vector group.Conclusion: β-catenin may play a pivotal role in occurrence and development of tongue squamous cell carcinoma.
