17β-雌二醇对胃癌细胞系SGC7901中NF-κB蛋白表达的影响

摘  要:目的:观察不同浓度雌二醇处理不同时间的情况下,人胃癌细胞系SGC7901中NF-B蛋白表达量的变化,探讨胃癌中雌激素对NF-B蛋白表达可能存在的影响.方法:SGC7901细胞经10 10mol/L和10 8mol/L的17-雌二醇(17β-estradiol,E2)处理6 h、12 h和24 h,应用Western Blot及灰度分析的方法检测蛋白表达量的改变,平均光密度值采用t检验分析.结果:E2作用于SGC7901细胞6h后,NF-κB蛋白表达量在10 10mol/L浓度组与对照组无明显差异,而10 8mol/L浓度组明显高于对照组(P<0.05).12h后,NF-κB蛋白表达量在10 10mol/L浓度组和10 8mol/L浓度组明显低于对照组(P<0.05).24 h后,NF-κB蛋白表达量在10 10mol/L浓度组和10 8mol/L浓度组低于对照组,但差异无统计学意义(P>0.05).结论:雌激素可对胃癌细胞系SGC7901中NF-κB蛋白表达量产生影响.Objective:To detect whether estrogen could regulate NF-κB protein expression in gastric cancer cell line SGC7901,NF-βB protein expressions of different concentrations of 17β-estradiol(E2) groups were detected at different time points.Methods:SGC7901 cell line was dealt with 10mol/L and 10mol/L E2 for 6 h,12 h and 24 h.Protein expression was tested by Western blot and image analysis methods.Average optical densities were processed with Student's test analysis.Results:After E2 treating for 6h,the protein expression of NF-κB was less in 10mol/L group than control group(P0.05).After E2 treating for 12 h,the protein expression of NF-κB were less in 10mol/L and 10mol/L groups than control group(P0.05).After E2 treating for 24 h,the protein expression of NF-κB were less in 10mol/L and 10mol/L groups than control group,but the difference did not had statistics significance(P0.05).Conclusion:17β-estradiol(E2) can influence NF-κB protein expression in gastric cancer cell line SGC7901.
