摘  要:目的观察大鼠全脑缺血再灌注损伤后海马区S期激酶相关蛋白2(S phase kinase-associated protein 2,Skp2)及其下游底物p27的表达变化。方法雄性Wistar大鼠60只随机分为2组:对照组(n=20),全脑缺血再灌注组(n=40)。采用Pulsinelli四血管结扎法建立大鼠全脑缺血再灌注损伤模型,全脑缺血30min后进行再灌注;对照组为假手术组,只分离血管。分别于大鼠全脑缺血再灌注4、7d用免疫组织化学法检测海马区星形胶质细胞和神经元变化;Western blot方法检测海马区Skp2及p27蛋白表达变化。结果与对照组相比,大鼠全脑缺血再灌注4、7d后,星形胶质细胞明显活化、增殖;海马CA1区神经元在再灌注7d时明显减少。Western blot检测结果显示,与对照组相比,在全脑缺血再灌注4d和7d海马区Skp2表达逐渐增高,而p27蛋白的表达逐渐降低(均P<0.05)。结论全脑缺血再灌注损伤大鼠海马区Skp2的表达升高,可能与缺血后神经元凋亡及胶质细胞活化有关。Objective To investigate the expression of S-phase kinase-associated protein 2(Skp2)and p27 in the hippocampus after reperfusion following forebrain ischemia in rats. Methods Sixty male Wistar rats were randomly divided into two groups:control group(n=20),ischemia-reperfusion group(n=40).The rats in ischemia-reperfusion group were induced by pulsinelli-4-vessel occlusion method,and the carotid clasps were removed after 30 min of 4-vessel occlusion and restoration of carotid blood flow.The rats in control group received sham operation.We investigated the change of neurons and astrocytes in hippocampus by immunohistochemistry 4 and 7 days after reperfusion.The expression levels of Skp2 and p27 were detected by using Western blot at the same time. Results The number of neurons in rat hippocampus was decreased,while astrocytes increased significantly after the ischemia-reperfusion injury.Astrocytes were activated obviously in the rat brain after reperfusion.The expression levels of Skp2 were increased markedly after 4 and 7 days of reperfusion,while those of p27 decreased after 4 and 7 days of reperfusion as compared with those in control group. Conclusion Skp2 was increased in the hippocampus after reperfusion following forebrain ischemia in rats,which may be associated with apoptosis of neurons and proliferation of astrocytes after cerebral ischemia-reperfusion injury.
