5-氟尿嘧啶联合奥沙利铂对SW480细胞成瘤性及其Galectin-3表达的影响

摘　　要：目的探讨5-氟尿嘧啶(5-Fu)联合奥沙利铂对SW480细胞成瘤性及其对肿瘤中Galectin-3(半乳糖凝集素-3)表达的影响。方法以人结肠癌SW480细胞建立裸鼠移植瘤模型,实验分5-Fu组,奥沙利铂组,5-Fu+奥沙利铂组和注射等体积生理盐水的对照组。采用免疫组织化学方法检测移植瘤中Galectin-3的表达,以图像信号采集与分析系统进行图像扫描分析。结果 5-Fu组、奥沙利铂组和5-Fu+奥沙利铂组抑瘤率分别为30.53%、34.15%和67.22%;免疫组化显示给药组肿瘤组织中Galectin-3表达明显减弱,Galectin-3在联合用药组肿瘤组织中表达(28.875±7.491)较对照组(281.438±45.639)和单独用药组(157.644±33.721,150.682±25.539)明显减弱,图像灰度值统计分析显示差异均具有统计学意义(P<0.05或P<0.01)。结论 5-Fu和奥沙利铂可抑制肿瘤细胞生长,二者联用抑瘤作用增强。肿瘤的生长体积与Galectin-3的表达具有同向性,提示两者之间可能存在直接或间接的联系。Objective To study the effects of 5-Fu combined with Oxaliplatin on tumorigenesis of SW480 cells and the expression of Galectin-3 in the tumor tissues. Methods Human colon cancer xenografts in nude mice were established by using SW480 cells.5-Fu group,Oxaliplatin group,5-Fu＋Oxaliplatin group,and saline control group were set up.By using immunohistochemistry the expression of Galectin-3 was detected in the xenografts.Image signal acquisition and analysis system was applied for image scanning analysis. Results The rate of tumor suppressor in 5-Fu group,Oxaliplatin group and 5-Fu＋Oxaliplatin group was 30.53%,34.15% and 67.22% respectively.Immunohistochemistry revealed that the Galectin-3 expression in 5-Fu＋Oxaliplatin group（28.875±7.491） was significantly reduced as compared with saline control group（281.438±45.639）,5-Fu group（157.644±33.721）and Oxaliplatin group（150.682±22.539）（P0.05 or P0.01）. Conclusion 5-Fu and Oxaliplatin could inhibit the growth of SW480 cells with a synergic effect.The Galectin-3 expression is decreased significantly in 5-Fu＋Oxaliplatin group,indicating Galectin-3 protein may play an important role in tumor cells growth.
