摘  要:目的检测Akt/mTOR信号通路中Akt、mTOR蛋白的活性形式(p-Akt、p-mTOR)在胃癌组织中的表达情况,并探讨其临床意义。方法采用免疫组织化学方法检测p-Akt、p-mTOR在93例胃癌组织及40例癌旁组织中的表达情况,结合患者临床病理资料进行分析。结果胃癌组织中,p-Akt、p-mTOR阳性表达者分别为51例(54.8%)和53例(57.0%),均显著高于癌旁组织中的表达,差异有统计学意义(P<0.01)。不同分化程度、侵犯深度、淋巴结转移、TNM分期的胃癌组织中,这两种蛋白的阳性表达率间差异均有统计学意义(P<0.05),而不同年龄、性别的胃癌组织中的这两种蛋白的表达间差异无统计学意义(P>0.05)。胃癌组织中p-Akt蛋白表达与p-mTOR蛋白表达呈正相关(r=0.327,P<0.01)。结论 Akt/mTOR信号通路蛋白在胃癌组织中的阳性表达与肿瘤进展相关,可能成为预测胃癌转移的新分子标志物和治疗靶点。Objective This study aims to examine the expression of active forms of the Akt/mTOR signal pathway proteins,p-Akt and p-mTOR,in gastric carcinoma and to analyze the correlations with clinical pathology characteristics.Methods The expression of p-Akt and p-mTOR was determined in 93 specimens of gastric carcinomas and 40 of control tissues by immunohistochemistry.The correlations between expression levels and clinical pathology features of gastric carcinoma were analyzed.Results The positive rates of p-Akt and p-mTOR in gastric carcinomas were 54.8%(51 cases) and 57.0%(53 cases) respectively,both significantly higher than in the control tissues(P0.01).The positive rates of the two proteins were also significantly different among cases of different depth of invasion,degrees of differentiation,lymph node metastasis status and clinical stages(P0.05);but were not significantly different among cases of different ages and genders(P0.05).The expression of p-Akt and p-mTOR was positively correlated in gastric carcinoma tissues(r=0.327,P0.01).Conclusion The expression of Akt/mTOR signal pathway proteins in gastric carcinoma is positively correlated with tumor progression,the detection of which shed light on the prediction of prognosis.Still,these proteins might serve as new molecular markers and targets for treatment.
