PSCA基因多态性与青海地区藏族胃癌遗传易感性的初步研究

摘　　要：目的初步研究PSCA基因多态性与青海地区藏族胃癌遗传易感性。方法收集60例藏族胃癌患者与60例健康人群的外周血标本,提取基因组DNA,采用dHPLC方法进行PSCA基因rs2976392位点分型。结果 PSCA基因rs2976392位点3种基因型G/A、A/A、G/G在胃癌病例组中的频率分别为51.67%、8.33%和40.00%,而在对照组中分别为43.33%、8.33%和48.33%。与G/G型比较,携带G/A型基因型者胃癌发生的危险性较高,OR=1.21(95%CI=2.21-3.63)。结论 PSCA基因rs2976392位点多态性可能与青海地区藏族人群胃癌易感性相关。Objective To investigate the polymorphisms of PSCA gene allele rs2976392 and the relationship between the genotype of PSCA and susceptibility to Tibetan gastric cancer in Qinghai area.Methods Genotypes were determined by PCR-based denaturing high-performance liquid chromatography analysis and direct DNA sequencing in 60 Tibetan patients with gastric cancer and 60 controls recruited in Qinghai area.Results In patients and normal controls,the frequency of G/A was 51.67%31（31/60） and 43.33%（26/60）,respectively,AA was 8.33%（5/60） and 8.33%（5/60） respectively,GG was 40.00%（24/60） and 48.33%（29/60）,respectively.PSCA G/A genotype were at increased risk of gastric cancer with an odds ratio of 1.21 for G/G homozygote（95%CI=2.21-3.63）.Conclusion The polymorphism of PSCA gene allele rs2976392 is associated with the susceptibility of Tibetan gastric cancer in Qinghai area.
