摘 要:目的研究肿瘤热休克蛋白70(HSP70)与灵芝孢子粉联合运用对荷瘤小鼠的治疗作用。方法液相色谱法纯化小鼠肝癌细胞株中的HSP70,运用SDS-PAGE及Western blot对纯化产物进行定性分析,再用毛细管电泳鉴定其浓度。通过动物实验,观察HSP70与灵芝孢子粉单独应用及联合应用的抗肿瘤作用。ELISA法测免疫小鼠血清中的IFN-γ、CXCL-9、CXCL-10的水平。结果 HSP70与灵芝孢子粉联合应用较单独应用的治疗效果更明显。单独应用灵芝孢子粉能延长荷瘤小鼠的生存期限,其中1只获得长期带瘤生存,而HSP70可使40%荷瘤小鼠的肿瘤全部消退,获得长期无瘤生存,与对照组相比较差异显著(P<0.01)。联合应用组40%荷瘤小鼠的肿瘤完全消退,3只小鼠获得长期生存,其中2只为无瘤生存,且肿瘤体积及质量均较分别单独应用组差别显著(P<0.05或P<0.01),平均生存期>(84.6±7.8)d,与对照组相比差异显著(P<0.01)。两药单独应用组、联合应用组血清IFN-γ、CXCL-9、CXCL-10水平均显著高于对照组(P<0.05或P<0.01)。联合应用组血清IFN-γ、CXCL-9、CXCL-10水平较单独应用组差别显著(P<0.05或P<0.01)。结论 HSP70与灵芝孢子粉联合应用对荷瘤小鼠有明显的治疗作用,可明显抑制肿瘤进展,提高生存率。以上结果对研究人类恶性肿瘤的免疫治疗具有较为重要的借鉴意义。Objective To investigate the therapeutic efficacy of the combination of tumor-derived HSP70 and ganoderma lucidum spores on tumor-bearing mice.Methods HSP70 was purified from the murine tumor cell line by liquid chromatograpy.And then identified by SDS-PAGE and Western blot;the purity of purified product was analysed by capillary electrophoresis.The anti-tumor efficacies of mono-administration of HSP70 or ganoderma lucidum spores and their combination were observed by zoopery.IFN-γ,CXCL-9 and CXCL-10 level in serum were assessed by ELISA.Results Combination of HSP70 and ganoderma lucidum spores was more effective than either of the two therapeutic agents alone in treating tumor-bearing mice.The average life span of mice treated with ganoderma lucidum spores was prolonged with one mice achieved long term survival.While 40% of the mice treated with HSP70,tumor disappeared eventually and their survival time was more than 90 days.HSP70 plus ganoderma lucidum spores could get tumors disappeared completely in 40% of mice,and 60% of mice achieved long term survival,the average life time of this group was over 84.6±7.8d,and the weight and the volume of tumors of this group was significantly smaller than that of the two agents applied alone.The serum levels of IFN-γ,CXCL-9 and CXCL-10 in the three treatment group were significantly higher than that of the control group.And the serum levels of three cytokines above in the combination group were remarkable promoted than that of the two agents applied alone.Conclusion The combination of HSP70and ganoderma lucidum spores displayed apparently therapeutic effect on tumor-bearing mice.This results provide significant information for the study of immunotherapy of human cancers.
