摘  要:目的观察玻璃体腔注射鼠神经生长因子对糖尿病大鼠视网膜结构的影响。方法选取SD雄性大鼠36只,随机分为正常对照组(A组)12只和糖尿病组24只,糖尿病动物模型通过链脲佐菌素60mg.kg-1左下腹腔一次性注射诱导,模型成功后随机分为玻璃体腔生理盐水注射组(B组)12只和鼠神经生长因子注射组(C组)12只,B组玻璃体腔注射生理盐水每次4μL,每周1次;C组注射鼠神经生长因子每次4μL(0.5g.L-1),每周1次。A组、B组和C组分别在4周和8周时随机选取3只(6眼)处死摘除眼球做石蜡切片观察视网膜光镜结构及3只(6眼)做电镜标本观察视网膜超微结构。结果各组大鼠血糖值:4周时A组(6.42±0.43)mmol.L-1、B组(30.07±3.62)mmol.L-1、C组(29.34±4.56)mmol.L-1,A组与B、C组之间差异均有统计学意义(均为P<0.05),B组与C组之间差异无统计学意义(P>0.05);8周时A组(5.78±0.47)mmol.L-1、B组(29.81±5.58)mmol.L-1、C组(31.26±4.03)mmol.L-1,A组与B、C组之间差异均有统计学意义(均为P<0.05),B组与C组之间差异无统计学意义(P>0.05)。视网膜光镜观察结果:A组视网膜各层结构清晰,排列整齐;B组4周时,视网膜结构改变不明显,8周时视网膜变薄,内外颗粒层变薄疏松,排列紊乱,神经节细胞减少;C组4周、8周时无明显改变。视网膜电镜观察结果:B组4周时视细胞外节膜盘模糊不清,膜盘间隙扩大,8周时视细胞膜盘间隙进一步扩大,排列紊乱,局部断裂,线粒体肿胀,并可见空泡变;C组较B组结构规则,4周时未见明显改变,8周时外节膜盘部分模糊,排列尚规则,间隙轻度扩大。结论玻璃体腔注射鼠神经生长因子对糖尿病大鼠视网膜结构具有明显保护作用。Objective To determine the effect of intravitreal administrated mouse nerve growth factor on retinal structure of diabetic rats.Methods Thirty-six SD rats were randomly divided into normal control group(group A,12 rats) and experiment group(24 rats).Experiment group was induced diabetic animal model with 60 mg·kg-1 of streptozotocin through intraperitoneal administration.After the models were established successfully,the rats were randomly divided into intravitreal administrated physiological saline group(group B) and intravitreal administrated nerve growth factor group(group C),12 rats in each group.The rats in group B received intravitreal administrated physiological saline with 4 μL per eye,once a week,and the rats in group C received intravitreal administrated 0.5 g·L-1 mouse nerve growth factor with 4 μL per eye,once a week.The eyeballs were randomly executed in 3 rats(6 eyes) of each group for paraffin section and observed under optical microscope and 3 rats(6 eyes) for observation under transmission electronic microscope at 4 weeks and 8 weeks,respectively.Results The blood glucose level of rats in group A,group B and group C at 4 weeks were(6.42±0.43)mmol·L-1,(30.07±3.62)mmol·L-1,(29.34±4.56)mmol·L-1,there were statistical differences between group A and group B or group C(all P0.05),but there was no statistical difference between group B and group C(P0.05);Blood glucose levels at 8 weeks were(5.78±0.47)mmol·L-1,(29.81±5.58)mmol·L-1,(31.26±4.03)mmol·L-1,there were statistical differences between group A and group B or group C(all P0.05),but there was statistical difference between group B and group C(P0.05).Optical microscope observation:The structure of each retinal layer in group A was clear and arranged orderly;In group B,there was no remarkable changes at 4 weeks,and the retinal thickness was thin,inner and outer nuclear layer puffed and arranged disorderly,the ganglion cell decreased at 8 weeks;In group C,there was no c
