摘 要:目的:探讨2种不同卡铂用药方式对人舌鳞状细胞癌株Tca8113耐药性的影响。方法:将Tca8113细胞株随机设置为1个对照组、6个不同程度的用药组和1个大剂量冲击用药组,然后加入相应浓度卡铂体外作用。用MTT法检测每组细胞的IC50值(半数抑制浓度),RT-PCR法检测每组细胞ERCC1 mRNA的表达。结果:随着药物浓度和作用次数的增加,小剂量逐步耐药法诱导的各组细胞IC50值和ERCC1 mRNA表达量均呈逐步递增的趋势,且它们呈正相关(r=0.878,P<0.01);大剂量冲击用药组细胞IC50值和ERCC1 mRNA表达量界于小剂量逐步耐药法的较小剂量组和较大剂量组之间,且有统计学差异(P<0.05)。结论:小剂量逐步多次用药法易引起Tca8113对卡铂的耐药性。Objective: To investigate the effect of two different methods of carboplatin administration on the drug- resistance of human tongue cancer Tca8113 cells in vitro. Methods: Tca8113 cells were randomly divided into a control group,six tested groups and a large dose shock group. Cells were exposed to the corresponding concentrations of carboplatin. The half inhibitory concentration (IC50) of each group was detected by MTT assay. The ERCC1 mRNA expression was detected by RT- PCR. Results: The value of IC50 and ERCC1 mRNA expression were gradually increased when the cells were exposed to increasing dose of carboplatin, furthermore, they were positively correlated(r=0.878, P0.01). The values of IC50 and ERCC1 mRNA of large dose shock group were between lower dose groups and higher dose groups(P0.05). Conclusion: Gradually increasing dose of carboplatin may induce drug- resistance of Tca8113 cells.
