摘 要:目的:探讨口腔癌中缺氧诱导因子(hypoxia-inducible factor-1α,HIF-1α)及肺耐药蛋白(LRP),DNA拓扑异构酶Ⅱ(TOPOⅡ),谷胱甘肽-s-转移酶-π(GST-π)的表达及其相关性。方法:采用免疫组化(SP法)技术检测57例口腔癌组织中HIF-1α、LRP、TOPOⅡ、GST-π蛋白的表达。结果:HIF-1α、LRP、TOPOⅡ及GST-π蛋白表达阳性率分别为63.16%、45.61%,38.60%和57.90%。HIF-1α的表达与淋巴结转移、临床分期相关,与LRP、GST-π的表达呈正相关关系,而与TOPOⅡ无明显相关性;LRP的表达与临床分期有关,GST-π与淋巴结转移有关。结论:口腔癌缺氧微环境通过HIF-1α影响肿瘤耐药相关蛋白的表达,联合检测为口腔癌的综合治疗方案的制定有临床意义。Objective:To investigate hypoxia-inducible factor-1α(HIF-1α) expression and its relation to drug resistance related proteins in oral carcinoma.Methods:Immunohistochemical S-P method was used to examine the expression of HIF-1α,lung resistance protein(LRP),topoisomeraseⅡ(TOPOⅡ) and glutathione-s-transferase-π(GST-π) in 57 cases of oral carcinoma.Results:The positive expression rates of HIF-1α,LRP,TOPOⅡ and GST-π in oral carcinoma were 63.16%,45.61%,38.60% and 57.90% respectively.The expression of HIF-1α was correlated with lymph node metastasis and clinical stage.There was significant positive correlation between the expression of HIF-1α and LRP or GST-π,but not with TOPOⅡ.The expression of LRP was related to clinical stage,and relation was found between GST-π and lymph node metastasis.Conclusion:Microenvironmental hypoxia may modulate the expressions of tumor drug resistance genes via HIF-1α.The expression detection of HIF-1α,LRP,TOPOⅡ and GST-π in oral carcinoma may be valuable for therapeutic regimen.
