大豆异黄酮对荷瘤去卵巢裸鼠肿瘤生长的影响

摘  要:目的探讨大豆异黄酮总提取物对荷瘤去卵巢鼠雌激素依赖型乳腺肿瘤的影响。方法以6.25,12.5和25 g/kg bw剂量大豆异黄酮总提取物喂食裸鼠2 w,去除卵巢并种植MCF-7肿瘤细胞,继续喂食16 w。用常规免疫组织化学方法检测肿瘤细胞核抗原Ki-67的表达,实时定量PCR(real-time PCR)检测雌激素相关基因pS2表达,化学发光酶免疫法检测裸鼠血清中雌激素的含量。结果与对照组比,喂食大豆异黄酮总提取物组裸鼠MCF-7移植瘤体积明显增加(P<0.05),肿瘤细胞增殖能力显著增强(P<0.05),pS2基因的表达增强(P<0.05),同时血清中雌激素的水平也因喂食大豆异黄酮总提取物而升高(P<0.05)。结论在本实验浓度范围内,青春期喂食大豆异黄酮总提取物对去卵巢裸鼠雌激素依赖型乳腺癌细胞移植肿瘤的生长有促进作用。Objective To explore the effects of soy isoflavone extracts on the growth of estrogen-dependent human breast cancer(MCF-7) tumors implanted in ovariectomized nude mice.Method The nude mice were fed with low,moderate and high doses of soy isoflavone extracts(6.25,12.5 and 25g/kg bw)respectively for 2 w before implanted with human breast cancer MCF-7 cells.The expression of Ki-67 and pS2 was detected by immunohistochemistry.and real-time PCR respectively.The estrogen level in serum was measured by chemiluminescence enzyme immunoassay.Results Compared with the control group,dietary soy isoflavone extracts had significant stimulatory effect on MCF-7 tumor growth in mice(P 0.05).The Ki-67 and pS2 mRNA expression in tumors was significantly increased by 6.25g/kg and 12.5g/kg dose of soy isoflavone extracts(P0.05).The estrogen level in serum was higher than that in control group(P 0.05).Conclusion Soy isoflavone extract has stimulatory effect on tumor growth.
