摘  要:目的观察人类快速延迟整流性钾通道基因表达的钾离子通道蛋白(HERG1)和血管内皮生长因子(VEGF)在胃癌组织中的表达,探讨两者与胃癌预后的关系。方法应用免疫组织化学方法(SP法)、逆转录-聚合酶链反应(RT—PCR)技术方法对80例胃癌组织标本及对应正常胃黏膜组织中的HERG1蛋白和VEGF的表达进行检测,并进行回顾性随访。结果免疫组织化学、RT—PCR检测胃癌组织中HERGl的阳性表达率分别为60%、90%,VEGF的阳性表达率分别为55%、85%;HERG1蛋白表达与淋巴结转移,远处转移关系密切;VEGF的表达与胃癌的浸润程度、TNM分期、分化程度、淋巴结转移明显相关(P〈0.05),而与性别、肿瘤大小、部位无明显相关。结论HERG1蛋白和VEGF的过度表达与胃癌的发生发展有关,在肿瘤侵袭、转移过程中起重要作用,其共位表达有助于判断胃癌预后。Objective To investigate the expression of HERG1 protein and vascular endothelial growth factor (VEGF) in gastric carcinomas and the correlation between HERG K^+ channels and VEGF. Methods Reverse transcription and PCR (RT-PCR) and immunohistochemistry (IHC) were used to detect the expression of HERG1 gene and VEGF in 80 gastric carcinomas and the patients were followed-up. Results The positive expression of HERG1 and VEGF was detected in 48 cases (60%) and 44 cases (55%) by immunohistochemistry, and in 72 cases (90%) and 70 cases (85%) by RT-PCR, respectively. There was a significant correlation between the positive expression of VEGF and depth of invasion, TNM stage, differention, lymphatic and distant matastasis in gastric carcinomas (P 〈 0.05 ). The positive expression of HERG1 was correlated with lymphatic and distant metastasis. Conclusion HERG K^+ channels and VEGF overexpression in gastric carcinomas appeared to be correlated with tumor genesis and development, and may play an important role in the tumor invasion and metastasis. The co-expression of HERG K + channels and VEGF could be well-characterized indicator to prognosis of gastric cancer.
