肿瘤相关巨噬细胞对大肠癌细胞株HT29迁移和增殖的影响

摘　　要：目的观察巨噬细胞在大肠癌中的作用。方法从外周血中分离单核细胞，分别用脂多糖（LPS）和白细胞介素（IL）-4刺激，将不同激活状态下的巨噬细胞与HT29细胞共培养，观察巨噬细胞对HT29细胞生物学特性的影响。结果经典激活的巨噬细胞诱导结肠癌细胞凋亡，凋亡细胞比例显著上升（17．93％比0．23％）；选择性激活的型巨噬细胞明显促进HT29细胞的增殖，S期与对照组比较差异有统计学意义（11．29％比5．61％）；迁移实验显示，选择性激活的巨噬细胞还能促进HT29细胞的迁移，通过Transwell的细胞数量大概是对照组的20倍。结论选择性激活的巨噬细胞能促进HT29细胞的增殖和迁移。Objective To observe the role of tumor associated macrophages （TAMs） in colorectal cancer. Methods Monocytes were isolated from peripheral blood, and stimulated with lipopolysaccharide （LPS） or interleukin （IL）-4 respectively. The macrophages and HT29 cells were co-cuhured. The impact of macrophages on biological characteristics of HT29 cells was observed. Results CaMφAaMφ induced apoptosis of HT29 cells and the proportion of apoptotic cells was obviously increased （ 17. 93% vs. 0. 23% ）. AaMφ promoted proliferation of HT29 cells proliferation, and as compared with the control group, proportion of S phase cells was evidently increased （11.29% vs. 5.61%）. Migration experiments showed AaMφalso promoted migration of HT29 ceils, and the number of migrating cells was nearly 20 times of the control group. Conclusion AaMφcan promotes proliferation and migration of HT29 ceils.
