摘　　要：目的从mRNA和蛋白水平检测原发性甲状腺癌中基质金属蛋白酶13(MMP13)基因的表达及其与病变的关系。方法采用逆转录聚合酶链式反应(RT-PCR)、蛋白印迹(Western blotting)和免疫组织化学技术检测62对配对甲状腺癌组织和癌旁正常组织中MMP13的表达情况。结果①RT-PCR显示,62例甲状腺癌组织中MMP13 mRNA的表达水平高于癌旁正常组织(P<0.05)。②Western blotting检测结果显示,MMP13在甲状腺未分化癌与分化型癌组织中的表达率高于其相对应的癌旁正常组织(P<0.05)。③免疫组织化学检测结果显示,62例甲状腺癌患者中的MMP-13总阳性率大于正常组织(P<0.05);未分化癌患者中的MMP13表达阳性率高于分化型癌,差异有显著性(P<0.05)。淋巴结转移阳性组的癌组织中MMP13的阳性表达率高于淋巴结转移阴性组(P<0.05)。TNM分期中,高分期组(Ⅲ、Ⅳ期)的癌组织中MMP13的阳性表达率较低分期组(Ⅰ、Ⅱ期)高,组间差异有显著性(P<0.05)。不同性别和年龄患者的MMP13阳性表达率差异无显著性(P>0.05)。结论 MMP13与甲状腺癌的组织类型和淋巴结转移密切相关,且与甲状腺癌进展密切相关,其可作为甲状腺癌病理改变参考因子和诊断的参考指标。[ Objective] To detect the matrix metalloproteinase 13 （MMP13） expression of primary thyroid tumor in mRNA and protein levels and the relationship with the disease. [Methods] MMPI3 expression in 62 pairs of matched thyroid carcinoma and adjacent normal tissue was detected by RT-PCR, western Blot and immunohistochemistry. [ Results ] ①RT-PCR showed that 62 cases of thyroid cancer MMP13 mRNA expression level was significantly higher than normal tissue （P 〈0.05）. ②Westem-blot test results showed that MMP13 in anaplastie thyroid cancer and differentiated cancer tissues was significantly higher than their corresponding adjacent normal tissues （P 〈0.05）. ③Immunohistoehemistry results showed that 62 eases of thyroid cancer, total positive rate of MMP-13 was higher than normal tissue （P 〈0.05）; Undifferentiated carcinoma MMP13 expression was higher in differentiated earcinoma, the difference was significant （P 〈0.05）. Thyroid carcinoma with lymph node metastasis group was significantly higher than the expression of MMP13 lymph node negative group （P 〈0.05）. TNM staging, MMP13 expression level of high-stage group （Ⅲ-IV stage） was higher than lowstage group （ I - Ⅱ period）, the difference between groups was significant （P 〈0.05）. There was no significant difference between MMP13 expression and patient gender and age （P 〈0.05）. [ Conclusion] MMP13 is closely related with type and lymph metastases of thyroid tumors andthyroid cancer progress, reflecting the pathological changes.
