摘 要:目的检测端粒重复序列结合因子1(TRF1)和端粒保护蛋白1(hPOT1)在鼻咽癌组织中的表达,探讨其在鼻咽癌发生过程中的作用及其临床意义。方法应用荧光定量RT-PCR检测47例鼻咽癌组织和10例正常鼻咽部黏膜组织中TRF1及hPOT1的mRNA表达水平。结果①鼻咽癌组织中TRF1 mRNA的表达明显低于正常鼻咽部黏膜组织(P=0.027),hPOT1的mRNA表达强度明显高于正常鼻咽部黏膜组织(P=0.013)。②TRF1 mRNA的表达与鼻咽癌T分期相关(P=0.038),但与临床分期和有无颈部淋巴结转移无明显相关(P>0.05)。③鼻咽癌组织中hPOT1 mRNA的表达与鼻咽癌临床分期、T分期和有无颈部淋巴结转移相关(P=0.013,P=0.025,P=0.041)。④鼻咽癌组织中TRF1 mRNA与hPOT1 mRNA的表达呈负相关(r=-0.461)。结论鼻咽癌组织中TRF1 mRNA的表达下降可能与鼻咽癌的发生相关,hPOT1 mRNA的表达增强可能与鼻咽癌的发生和发展相关。[Objective] To investigate the expression of TRF1 and hPOT1 and its correlation with the clinical pathological parameters in nasopharyngeal carcinoma, and to reveal the potential mechanisms of TRF1 and hPOT1 underlying nasopharyngeal carcinoma. [ Methods ] The technique of quantitative reverse transcription polymerase chain reaction was applied to detect the TRF1 and hPOT1 mRNA expression in 47 cases of nasopharyngeal carcinoma tissues and 10 cases of normal nasopharyngeal mucosa, mRNA expression level of TRF1 and hPOT1 in different groups was compared statistically. [Results] The relative mRNA expression of TRF1 in nasopharyngeal carcinoma tissues was lower than that in normal nasopharyngeal mucosa (P =0.027), the mRNA expression of hPOT1 in nasopharyngeal carcinoma dssues was higher than that in normal nasopharyngeal mucosa (P =0.013). Besides, the relative mRNA expression of TRF1 was associated with tumor size (P =0.038), but no association with clinical stages and lymphocyte metastasis. Moreover, the relative mRNA expression of hPOT1 was associated with tumor size, clinical stages and lymphocyte metastasis (P =0.013, P =0.025, P =0.041). Furthermore, TRF1 expression was inversely correlated with hPOT1 (r =-0.461). [ Conclusions] TRF1 expression is down-regulated in nasopharyngeal carcinoma might relates to tumor occurrence. Meanwhile, hPOT1 expression up-regulated in nasopharyngeal carcinoma might relates to tumor occurrence and development.
