摘 要:目的:观察红景天苷对乳鼠心肌细胞胞浆Ca2+浓度的影响并分析其可能的作用机制。方法:应用荧光指示剂Fluo-3/AM负载培养大鼠乳鼠的心肌细胞,用激光共聚焦显微镜动态观察胞内游离钙荧光信号强度的变化,检测不同浓度红景天苷对培养心肌细胞胞内游离钙离子浓度([Ca2+]i)的影响。结果:红景天苷浓度为15 mg/L、30 mg/L和60 mg/L时,细胞内的平均[Ca2+]i升高,峰值分别为574.08±4.65、591.86±3.64和618.66±4.27(均P<0.01);有剂量依赖性而无时间依赖性。用维拉帕米阻断细胞膜外钙内流时,红景天苷同样引起细胞内[Ca2+]i升高,峰值由357.74±3.13、387.17±2.37和391.43±1.34分别上升到480.86±3.98、496.70±3.08和522.18±3.19(均P<0.01)。结论:红景天苷能升高乳鼠心肌细胞中[Ca2+]i,其机制可能与其促进肌浆网钙离子释放有关。AIM:To observe the effects of salidroside on intracellular free Ca2+ concentration in cultured rat cardiomyocytes.METHODS: Primarily cultured cardiomyocytes of neonatal rats were divided into control group,different concentrations of salidroside groups and verapamil pretreatment+different concentrations of salidroside groups.The fluorescent intensity of intercellular free calcium concentration(i) in cultured cardiomyocytes of newborn rats loaded with fluo-3/AM(5 μmol/L) was measured by laser scanning confocal microscopy.RESULTS: Salidroside at concentrations of 15 mg/L,30 mg/L and 60 mg/L elevated i in cultured rat cardiomyocytes with the peak values of 574.08±4.65,591.86±3.64 and 618.66±4.27,respectively(all P〈0.01),indicating that the effect of salidroside on the level of i was dose-dependent.In the presence of verapamil in D-Hanks solution,salidroside also elevated the fluorescent intensity of i in cardiomyocytes from 357.74±3.13,387.17±2.37 and 391.43±1.34 to 480.86±3.98,496.70±3.08 and 522.18±3.19,respectively(all P〈0.01).CONCLUSION: Salidroside increases the release of i from sarcoplasmic reticulum in cultured rat cardiomyocytes.
