摘 要:目的检测多药耐药相关蛋白(MRP)和肺耐药蛋白(LRP)在肺癌组织及外周血中的表达及其相关性,探讨其临床意义。方法采用逆转录-聚合酶链反应(RT-PCR)检测47例患者肺癌组织及外周血的MRP、LRP mRNA的表达水平。结果肺癌组织及外周血,MRP mRNA的阳性表达率分别为74.5%(35/47)和70.2%(33/47),两者之间呈显著相关(r=0.686,P<0.01);LRP mRNA的阳性表达率分别为82.9%(39/47),76.6%(36/47),两者之间亦呈显著相关(r=0.368,P<0.01);肺癌组织MRP mRNA和LRP mRNA的表达表现为正相关(r=0.384,P<0.01)。结论肺癌患者MRP、LRP mRNA的表达密切相关,检测外周血中MRP、LRP mRNA表达能间接反映癌组织对化疗药物的耐受性。Objective To investigate the expression level and its clinical significance of multidrug resistance (MDR) markers (MRP mRNA and LRP mRNA) in lung cancer tissues and peripheral blood, and then to explore clinical sense of the result. Methods The expression of Lung cancer resistance protein (LRP), multidrug resistance protein (MRP) of MDR makers were detected in 47 cases of patient lung cancer tissues by reverse transcription and polymerase chain reaction (RT-PCR) technique. Results The positive expression rate of MRP mRNA in lung cancer tissues and peripheral blood was 74.6% (35/ 47) and 70.2% (33/47). A significant correlation was found between the two (r=0. 686, P〈0.01) ; The expression of LRP rnRNA was 82.9% (39/47) and 76.6% (36/47). It was significantly correlative between the two (r=0. 368, P〈0.01). Therefore, there were significant relationships between MRP mRNA and LRP mRNA expression in lung cancer tissues (r= 0. 384, P〈0.01). Conclusion The mRNA levels of MRP and LRP expression in the peripheral blood of lung cancer patients is closely related, as a result, detecting it can reflect the tolerance between the chemotherapeutic drug and lung cancer tissues indirectly.
