周期性牵张应力对C2C12成肌细胞凋亡的影响

摘　　要：目的探讨不同条件下的周期性牵张应力对C2C12成肌细胞凋亡的影响。方法利用细胞加力仪器与弹性加力板对体外培养C2C12成肌细胞施加不同频率、不同幅度的牵张应力。细胞计数、MTT实验观察细胞的活性,并利用Western Blot,DNA ladder,PARP剪切实验等方法检测在应力条件下成肌细胞凋亡规律。结果较低幅度的牵张应力(5%和10%),促进成肌细胞的增殖;随着牵张应力的幅度的增大,当拉伸条件为15%及20%时,细胞数目较对照组显著降低(P<0.05),DNA ladder实验和PAR P剪切实验发现随着拉伸幅度的增加,剪切型的PARP逐渐增多,DNA ladder显示凋亡特征的带型逐渐出现,并且随着幅度的增加,凋亡带逐渐明显。结论在较低幅度的牵张应力作用下,成肌细胞以增殖为主,随着应力的增大,细胞开始凋亡。临床医生牵张成骨治疗过程中,应注意合理施力。【Objective】To investigate the effect of cyclic stretch of different magnitude on the viability and apoptosis of C2C12 myoblast.【Method】Flexcell strain system was introduced and the surface of the 6-well collagen coated plates can extend.By this,cultured C2C12 myoblasts can be stretched by different stretch magnitudes.Cell counting and MTT was used to estimate the viability of the cells.PARP cleavage,western blotting and DNA ladder were introduced to test the apoptosis of the C2C12.【Results】Lower magnitude（5% and 10%） of stretch induced C2C12 cell proliferation.However,cell viability decreased when higher magnitude stretch（15% and 20%） was used.DNA ladder and western blotting of PARP showed obviously apoptosis featured band and cleavage PARP,suggesting apoptosis contributed at least partially to the observed cell viability.【Conclusion】Lower magnitude of stretch was prone to induce C2C12 myoblast proliferation.However,when the magnitude increased the cell viability decreased,apoptosis became more.Clinician should pay attention to the magnitude of stretch they apply when they do distraction osteogensis to the craniofacial system.
