重组人血管内皮抑素联合化疗不同用药时序对肺癌移植瘤生长和凋亡的影响

摘　　要：目的观察恩度与化疗联合应用的不同时序对裸鼠A549移植瘤生长和凋亡的影响。方法60只荷A549人肺癌细胞株移植瘤裸鼠随机分为实验组和对照组，实验组分为D1、D3、195、D7组，给予恩度治疗并分别在恩度治疗后第1、3、5、7天进行化疗，比较各组裸鼠移植瘤的生长情况和凋亡指数。结果治疗后第40天各治疗组移植瘤体积均明显小于对照组且差异有极显著性垆〈0．01），D3、D5、D7组肿瘤体积小于D1组（P〈0．05）D1、193、D5、D7组凋亡指数分别为（14．28±1．93）、（15．76±1．96）、（15．44±1．91）、（16．55±2．07），与对照组（2．45±0．68）相比差异有极显著性伊〈0．01），D3、D5、D7组凋亡指数均高于D1组但仅D7组与D1组比较差异有显著性（P〈0．05kD1、D3、D5、D7组抑瘤率分别为47．97％、46．78％、46．91％、53．52％。结论在恩度治疗后第3、5、7天给予化疗较恩度与化疗同时给药，有助于促进裸鼠移植瘤细胞凋亡及抑制其生长，恩度治疗后第7天可能是给予化疗的最佳时点。Objective To observe the growth and apoptosis effects of human lung carcinoma cell A549 treated in endostar combined with chemotherapy at different sequences. Methods 60 tumorbearing nude mice transplanted with human lung carcinoma cell A549 were randomly divided into experimental group and control group. And experimental group were divided into group D 1, D3, D5 and D7, given chemotherapy at 1ts, 3 th 5 th and 7 th day after recombinant human endostar treatment. The growth change and tumor apoptotic index were compared between the groups after the therapy finished. Results The tumor volume in treatment groups was significantly smaller than that of the control group at 40 ^th day after endostar treatment（P 〈 0.01）; the tumor volume of group D3, D5, D7 were smaller than that of the group D1 （P 〈 0.05）; the apoptotic index of group D1, D5, D3, D7 were （14.28 ± 1.93）,（15.76 ± 1. 96）, （15.44 ±1.91） and （16.55 ± 2.07） respectively, which were significantly different from those of the control group（AI：2.45 ± 0.68） （P 〈 0.001）; the apoptotic index of group D3, D5, D7 were higher than that of the group D1, but there were significant differences only between group D7 and group DI（P=0.011）; the tumor inhibitory rates of group D1, D3, D5, D7s were 47.97%, 46.78%, 46.91% and 53.52% respectively. Conclusion The effect of chemotherapy given at the 3 ^th, 5 ^th, 7 ^th day after treatment by endostar to inhibit tumor growth and improve tumor apoptotic index were more effective than those two were given in the same time. Chemotherapy given at the 7 ^th day after endostar treatment could be the best effective point.
