摘  要:目的:探讨骨髓间质分离的多能成体祖细胞(MAPCs)移植后在大鼠脑组织内神经细胞分化的形态学观察。方法:制作帕金森病(PD)大鼠模型,将在体外纯化、增殖和用5-溴-2脱氧尿苷(BrdUrd)处理过的MAPCs注入PD大鼠脑内。3个月后,对移植大鼠采用免疫组化技术、免疫电镜等方法鉴定和分析MAPCs在大鼠脑组织内神经元样细胞分化的形态学。结果:MAPCs在中脑黑质和纹状体区分化为神经元样细胞和多巴胺能神经元;电镜下观察到MAPCs所分化的神经细胞与其他神经细胞形成突触联系。结论:骨髓间质分离的MAPCs移植后能在大鼠脑组织微环境中自主分化为多巴胺能神经细胞并有效地与其他神经细胞形成突触联系。因此MAPCs有望成为中枢神经系统疾病自体移植治疗的最佳候选干细胞之一。Objective: To observe the morphology of the differentiation of the neurons cells in the brain tissues after transplantation of the multipotent adult progenitor cells(MAPCs) derived from bone marrow into rat brain.Methods: The rat models with Parkinson's disease were established and MAPCs prelabelled with bromodeoxyuridine(BrdUrd) were injected into left striatum and substantia nigra.Immuno-fluorescence and electron microscopy were used to evaluate and analyze that MAPCs differentiate into neuron-like cells after three months.Results: After implantation,The MAPC cell was capable of differentiating in vivo into dopamine-neurons in striatum and substantia nigra.The finding of synapse with other nerve cells in the electricon microscopy implicated MAPCs-derived neuron should have an important role in reconstruction of neural circuitry.Conclusion: The transplanted MAPCs can develop spontaneously into DA neurons and form synaptic with other nerve cells.MAPCs may provide a powerful autoplastic therapy for a variety of central nervous system disorders.
