摘 要:目的:观察高尿酸肾病模型大鼠血尿酸、肾脏组织病理学及肾组织中的TGF-β1、TNF-α的变化及中药复方对TGF-β1、TNF-α表达的调节作用。方法:采用高嘌呤饲料加腺嘌呤灌胃制作高尿酸血症肾病大鼠模型,并分组处理,中药治疗组用中药煎剂,别嘌呤醇治疗组用别嘌呤醇溶液,模型对照组及正常对照组给予蒸馏水灌胃,用药持续4周。观察血尿酸、肾脏组织病理学及肾组织中的TGF-β1、TNF-α的变化。同时,观察中药复方对上述各项指标的调节作用。结果:高尿酸血症肾病大鼠肾组织TGF-β1、TNF-α表达明显上调,且中药复方能调节其表达。结论:高尿酸血症肾病大鼠肾组织TGF-β1、TNF-α表达明显上调;中药有调节高尿酸血症肾病大鼠肾组织中TGF-β1、TNF-α表达的作用。Objective To observe the blood uric acid level,the changes of pathology and expressions of TGF-β1 and TNF-α in the kidney tissue of the rat model with hyperuricemic nephropathy,and explore the adjustment effects of Chinese herb on the expressions of TGF-β1 and TNF-α.Methods The rats were fed with high-purine diet and treated with adenine by intragastric administration to establish the rat model with hyperuricemic nephropathy.All rats were randomized into 4 groups:three model groups were treated with Chinese herb solution,allopurinol and distilled water respectively;and one normal control group was treated with distilled water.The drug intervention lasted 4 weeks.The blood acid uric level of the rat model with hyperuricemic nephropathy was measured,the changes of pathology and expressions of TGF-β1 and TNF-α in the kidney tissue were observed.And the adjustment effects of Chinese herb on the above indexes were also investigated.Results The blood uric acid level and the expression level of TGF-β1 and TNF-α in kidney tissue of the rat model with hyperuricemic nephropathy were increased significantly,comparing with normal control group(P 〈 0.01).Comparing with model group,the blood uric acid level and the expression level of TGF-β1 and TNF-α in kidney tissue of Chinese herb group were decreased obviously(P 〈 0.01).Conclusion The blood uric acid level and the expression level of TGF-β1 and TNF-α in kidney tissue of the rat model with hyperuricemic nephropathy were improved significantly,and the Chinese herb have the adjustment effects on the expressions of TGF-β1 and TNF-α in kidney tissue of the rat model with hyperuricemic nephropathy.
