摘  要:目的检测糖尿病大鼠睾丸组织中诱导型一氧化氮合酶(iNOS)及活性氧(ROS)的含量变化。方法构建雄性糖尿病大鼠模型,成功造模2个月时,取睾丸组织,分别应用实时荧光定量PCR、Westernblot检测正常对照组及糖尿病组大鼠睾丸组织内iNOSmRNA和蛋白的表达,用二氢乙啶活性氧探针检测ROS的含量。结果糖尿病组大鼠睾丸组织中iNOSmRNA和蛋白的表达及ROS的含量较正常对照组均明显增高(P〈0.05)。结论糖尿病组大鼠睾丸组织中iNOS及ROS的含量较正常对照组明显升高,iNOS与ROS可能参与了糖尿病睾丸组织的病理生理改变。Objective To explore the content of induced nitrogen monoxide synthase(iNOS) and reactive oxygen species(ROS) in testicular tissues of diabetic rats. Mothods Diabetic rat was constructed, after 2 months, the testicle was harvested for measurement of iNOS mRNA and protein expression with real-time PCR, Western blotting respectively, for measurement of ROS content with dihydroethidium probe. Results Expression of iNOS both at protein and mRNA were significantly increased in diabetic rats when compared to that of control rats (P〈0.05), ROS content was significantly increased in diabetic rats when compared to that of control rats(P〈0.05). Conclusion The results suggested that the expression of iNOS and ROS content significantly increased in testicular tissues of diabetic rats, iNOS and ROS might involve in the physiopathologic change of diabetic testicular tissues.
