胃癌组织中TGF-β1、TGF-βRⅡ、Smad2/3和CDC25蛋白的表达及其临床意义

摘　　要：目的检测TGF-β1、TGF-βRⅡ、Smad2/3及CDC25蛋白在胃癌组织中的表达并探讨它们在胃癌组织中的发生、发展的意义。方法利用免疫组织化学SP法检测110例胃癌、21例高级别上皮内瘤变、17例低级别上皮内瘤变、54例肠上皮化生、28例慢性萎缩性胃炎和57例正常胃黏膜组织中TGF-β1、TGF-βRⅡ、Smad2/3和CDC25蛋白的表达。结果 TGF-β1、TGF-βRⅡ、Smad2/3和CDC25在胃癌组织中的表达均高于正常胃黏膜组织。TGF-β1在胃癌组织中的表达高于上皮内瘤变、肠上皮化生和慢性萎缩性胃炎组织;Smad2/3在胃癌组织中的表达高于慢性萎缩性胃炎组织;CDC25在胃癌组织中的表达高于低级别上皮内瘤变组织,且在胃癌淋巴结转移组中的表达高于未转移组。TGF-β1与TGF-βRⅡ、Smad2/3和CDC25呈正相关,Smad2/3与CDC25也呈正相关。结论 TGF-β1、TGF-βRⅡ、Smad2/3和CDC25的表达是引起胃癌发生、发展的重要因素,可作为胃癌早期诊断的辅助指标。Purpose To investigate the expression of TGF-β1,TGF-βRⅡ,Smad2/3 and CDC25 in the carcinogenesis and development of gastric carcinoma.Methods The expressions of TGF-β1,TGF-βRⅡ,Smad2/3 and CDC25 were detected by immunohistochemistry in 110 of gastric carcinoma,21 of high-grade intraepithelial neoplasia,17 of low-grade intraepithelial neoplasia,54 of intestinal metaplasia,28 of chronic atrophic gastritis and 57 of normal gastric tissues.Results The expressions of TGF-β1,TGF-βRⅡ,Smad2/3 and CDC25 in the gastric carcinoma were higher than in normal gastric tissues.In gastric carcinoma,the expression of TGF-β1 was higher than in high-grade intraepithelial neoplasia,intestinal metaplasia and chronic atrophic gastritis.The expression of Smad2/3 in gastric carcinoma was higher than in chronic gastritis.The expression of CDC25 in gastric carcinoma was higher than in low-grade intraepithelial neoplasia,and CDC25 expression in gastric carcinoma with lymph node metastasis was higher than in without metastasis.TGF-β1 was positive correlated with TGF-βRⅡ,Smad2/3 and CDC25,respectively.And there was a positive correlation between Smad2/3 and CDC25.Conclusion The expressions of TGF-β1,TGF-βRⅡ,Smad2/3 and CDC25 might play an important role in carcinogenesis and development of gastric carcinoma,and these four proteins might be a helpful marker for the early diagnosis of gastric carcinoma.
