摘 要:目的:观察毛蕊异黄酮在体外对雌激素受体阳性细胞人乳腺癌MCF-7细胞的作用及其机制。方法:用不同浓度毛蕊异黄酮作用MCF-7细胞,并设未处理阴性对照组及17-雌二醇(17-estradiol,E2)阳性对照组。MTT法测定细胞增殖抑制的情况,FCM检测细胞凋亡率影响,并通过RT-PCR和免疫细胞化学法分别测定细胞中Bcl-2、Bax的mRNA和蛋白水平变化。结果:低浓度的毛蕊异黄酮(2、4和8μmol/L)促进MCF-7细胞增殖,而高浓度毛蕊异黄酮(16和32μmol/L)抑制MCF-7细胞增殖,并且呈剂量依赖性。同时,低浓度的毛蕊异黄酮能减少MCF-7细胞凋亡率(P<0.05),并且显著降低Bax表达水平,增加Bcl-2表达水平(P<0.05)。结论:毛蕊异黄酮在低浓度时能促进MCF-7细胞增殖,减少细胞凋亡,其机制可能与毛蕊异黄酮的雌激素作用有关。Objective:To investigate the effect of calycosin on estrogen receptor-positive human breast cancer MCF-7 cells in vitro,and to elucidate the posssible mechanism.Methods:Human breast cancer MCF-7 cells were treated with different concentrations of calycosin.The untreated group and 17-estradiol(E2)-positive group were used as the controls.Then cell proliferation was detected by MTT assay,the apoptosis rate was examined by flow cytometry,and the mRNA and protein levels of Bcl-2 and Bax were determined by reverse transcription-polymerase chain reaction(RT-PCR) and immunocytochemistry,respectively.Results:Calycosin at the low concentrations of 2,4 and 8 μmol/L stimulated the proliferation of MCF-7 cells compared with solvent control,whereas calycosin at the concentrations of 16 and 32 μmol/L inhibited cell proliferation in a dose-dependent manner.Calycosin at the low concentrations of 2,4 and 8 μmol/L decreased the percentage of early apoptosis in MCF-7 cells(P〈0.05),down-regulated mRNA and protein levels of Bax and up-regulated those of Bcl-2(P〈0.05).Conclusion:Calycosin at low concentrations can promote the proliferation of MCF-7 cells and inhibit the apoptosis,which may be due to its estrogenic action.
