摘  要:目的探讨针灸预处理对老年性痴呆模型大鼠GSK-3β和PP2A影响,揭示针灸防治AD的机制。方法建立AD大鼠模型,随机分为正常组、假手术组、模型组、预艾灸组、预电针组、预电针加灸组,通过酶联免疫法检测蛋白质磷酸酶2A(PP2A)和糖原合酶激酶-3β[(GSK-3β)]的活性。结果模型组GSK-3β的活性比正常组亦明显升高(P<0.01),而PP2A的活性比正常组和假手术组明显降低。预电针和预艾灸均可起到调节作用。结论针灸防治老年性痴呆的作用机制可能是通过调节Wnt信号转导通路,影响了学习记忆相关酶在海马区的表达而发挥作用的。Objective To discuss the effect of acupuncture and moxbustion pretreatment on GSK-3β PP2A,so to research the mechanism in treating and preventing AD disease. Methods To establish the AD model.All rats were divided into six groups at random: sham group,model group,moxibustion group,electro-acupuncture groups,electro-acupuncture and moxibustion group,ELISA method was used to detect the activity of PP2A and GSK-3β. Results The activity of GSK-3β in model group was increased significantly as compare to normal group(P0.01),but the activity of PP2A was decreased obviously.Electro-acupuncture and moxibustion could adjust those changes. Conclusion The adjusting mechanism of acupuncture to prevent and cure dementia is likely to regulatre the Wnt signal transduction pathways and affect the relevant enzyme expression of learning and memory in hippocampus.
