摘 要:目的探讨ApoE基因敲除小鼠主动脉组织中CD55、CD59的表达变化及意义。方法实验动物分为2组:对照组(C57BL/6小鼠10只)和动脉粥样硬化(AS)组(ApoE基因敲除小10只)。普通饲料喂养8周。监测体质量、血脂,观察斑块病变情况;采用免疫荧光和Real-ti me PCR技术,从蛋白水平和mRNA水平来观察CD55和CD59的表达变化。结果与对照组相比,AS组体质量增加[(27.14±1.60)g vs(29.50±1.38)g,P<0.05],总胆固醇和低密度脂蛋白明显增高〔(2.52±0.16)mmol/Lvs(12.74±1.79)mmol/L,(0.28±0.03)mmol/Lvs(9.06±1.13)mmol/L,P均<0.01〕;小鼠血清淀粉样蛋白-a水平有增加趋势[(5.08±2.34)ug/mLvs(7.06±4.81)ug/mL,P>0.05];AS组可见明显动脉粥样硬化斑块形成。免疫荧光显示,AS组CD55和CD59表达增强。Real-ti me PCR结果显示,与对照组比较,AS组CD55和CD59 mRNA相对表达量均有上升趋势,但差异无统计学意义(0.75±0.19 vs 4.20±0.45,1.21±0.21 vs 5.58±2.53,P均>0.05)。结论 ApoE基因敲除小鼠主动脉组织中CD55、CD59蛋白水平和mRNA水平表达上调,可能参与了动脉粥样硬化的发生和发展。Objective To investigate the levels of CD55 and CD59 expression on aortas in atherosclerosis.Methods Twenty male mice were obtained at 6 weeks of age.After 1 week of acclimatization,animals were divided into two groups: control group(C57BL/6 mice,n=10),and atherosclerosis(AS)group(ApoE-knock out mice,n=10).After 8 weeks of treatment with rodent chow,venous bloods were collected for lipids analysis,and aorta were prepared for atherosclerotic plaques,immunofluorescence and Real-time PCR.Results There were significantly differences in body weigh,lipids and atherosclerotic plaques between control group and AS group.Immunofluorescence showed that compared with control group,CD55 and CD59 expressions increased in AS group.Real-time PCR showed that the levels of CD55 and CD59 mRNA expressions showed a trend towards increased in AS group(0.75±0.19 vs 4.20±0.45,1.21±0.21 vs 5.58±2.53,P0.05).Conclusion The levels of CD55 and CD59 expression on aortas in ApoE-knock out mice were increased,and might play an important role in atherosclerosis.
