正交法优化马铃薯渣作培养代料培育凤尾菇及其菌糠中β-葡聚糖酶和植酸酶活性的检测研究

摘  要:使用正交法优化了马铃薯渣作为培养基代料的凤尾菇培育方法,得出了温度为20℃、马铃薯渣加入量为8%、棉籽壳加入量为70%、pH为7.0最优培育条件,于此条件下实际收获凤尾菇172 g。对菇体采摘后的菌糠进行了2种酶活检测,β-葡聚糖酶活性为0.0260 U/g,植酸酶活性在pH2.5下为0.0822 U/g,在pH5.2下则为0.0786 U/g,活性均偏低,不宜提取,但可在其用作饲料加工时相应减少2种酶添加量,以降低成本。该法在提高凤尾菇产量的同时合理利用了马铃薯渣,并为菌糠酶的开发利用做了探索研究。The main purpose of this work was to optimize the cultivated method of Pleurotus by orthogonal test, which used potato processing waste. The optimal conditions of cultivation were obtained by orthogonal test as follows: cultivation with 8% potato processing waste and 70% cotton seed hulls in culture medium, at pH 7.0, 20 %. The actual output of Pleurotus was 172 g. Then checked the biological activity of β-glucanase and phytase. The β-glucanase's activity of Pleurotus's mushroom residue was 0.0260 U/g, the phytase's activity at pH 2.5 and pH 5.2 were 0.0822 U/g and 0.0786 U/g. Although the activity was too low to extract, the result shows that we could add a bit of β-glucanase and phytase when we used mushroom residue as feedingstuff material. This work was not only enhanced the output of Pleurotus, but also utilized potato processing waste effectively. It dose an exploratory study of utilizating the enzyme in mushroom residue.
