摘  要:目的:测定正常人视网膜不同部位SOD、MDA、GSH、GST水平,探讨视网膜不同部位抗氧化能力的差异性,为相关眼病的发病机理及防治提供实验及理论依据。方法:取成人眼球,6人(12只眼),生理盐水冲洗,赤道部切开眼球,弃去眼前节及玻璃体,取黄斑部、后极部、周边部全层视网膜三组,制成10%组织匀浆作生物化学检验。分别应用黄嘌呤氧化酶法测定SOD,硫代巴比妥酸(TBA)法测定MDA,二硫代二硝基苯甲酸(DTNB)法测定GSH,化学比色法测定GST。结果:黄斑部、后极部、周边部视网膜SOD活力依次升高,GSH含量及GST活力依次升高,黄斑部最低,比较差异具有统计学意义(p<0.05);MDA含量依次降低,黄斑部最高,比较差异具有统计学意义(p<0.05)。结论:视网膜黄斑部、后极部、周边部三个部位的氧化指标即SOD、GSH、GST、MDA存在差异,从氧化反应角度为视网膜不同部位生理功能和疾病的发生提供理论依据。Objective:To estimate the antioxidant properties in different parts of retina through determining the levels of SOD,MDA,GSH,GST in order to investigate pathological mechanism and provide the possible avenue to treat the ophthalmological diseases caused by oxidative stress.Methods:The eyes of 6 adults(who were subjected to sudden death) were removed and washed with sterilized saline.Incisions were made at the equatorial plane of eyeballs,and the parts of full retina,including yellow spot(Y),posterior part(PO) and peripheral part(PE) were removed and homogenized for biochemical analysis,respectively.The levels of SOD were measured with xanthine oxidase method,MDA was measured with TBA,GSH with DTNB and GST with colorimetric method.Results: The levels of SOD,GSH and GST in yellow spot,posterior part and peripheral part of retina were revealed in the pattern of YPOPE(p 0.05),while the levels of MDA showed the pattern of YPOPE(p 0.05).Conclusions: Yellow spot,posterior part and peripheral part of retina have different antioxidant properties,which indicates the various vulnerabilities of the three parts of retina to oxidative stress.
