摘 要:目的探索表皮生长因子(EGF)培养体外人视网膜色素上皮细胞(hRPE)的最佳浓度和最佳作用时间。方法用CCK-8法观察EGF对体外hRPE增殖调控的剂量-效应关系、时间-效应关系。结果在相同浓度EGF作用下,hRPE吸光度值(A值)随作用时间延长而增加;在相同培养时间条件下,hRPE A值随EGF浓度增加而增加,EGF质量浓度<8 ng/mL的各相邻浓度实验组组间A值比较差异有统计学意义(P<0.05);EGF质量浓度≥8 ng/mL的各相邻浓度实验组组间A值比较差异无统计学意义(P>0.05)。结论 EGF对体外培养hRPE增殖调控存在剂量-效应关系和时间-效应关系,EGF的促增殖作用在8 ng/mL以上逐渐达到饱和。Objective To explore the best concentration and acting time of the epidermal growth factor(EGF)on the proliferation of human retinal pigment epithelial(hRPE) cells in vitro.Methods CCK-8 assay was adopted to identify the dose-response and time-effect relationship of the EGF on hRPE cells.Results Under the same concentration of EGF,the hRPE absorbance(A value) increased with the prolongation of time.During the same time,the hRPE absorbance increased with the rise of EGF concentration.During the same time,the difference between the two groups with adjacent concentrations was statistically significant when the EGF concentration below 8 ng/mL(P〈0.05),but the difference was no statistic significance when the EGF concentration above 8 ng/mL(P〉0.05).Conclusion The effect of EGF on the proliferation of hRPE cells is associated with the dose-response and time-effect relationship.The stimulation effect of EGF is gradually saturated when its concentration is above 8 ng/mL
