拉莫三嗪对戊四唑点燃大鼠模型行为学及抗氧化指标的影响

摘  要:目的探讨拉莫三嗪(LTG)在戊四唑(PTZ)点燃大鼠模型的作用。方法大鼠腹腔注射35 mg.kg-1PTZ建立癫痫点燃模型,取正常大鼠10只及造模成功大鼠30只分为4组,即正常对照组、PTZ模型组、LTG组(25mg.kg-1)、卡马西平阳性对照组(60 mg.kg-1),分别腹腔注射对应药物28天后,观察其行为学变化,28至31天检测Morris水迷宫行为学变化,第32天测定脑组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH)、过氧化氢酶(CAT)、丙二醛(MDA)含量。结果拉莫三嗪可显著延长大鼠癫痫发作的潜伏期、降低发作分级和持续时间;缩短逃避潜伏期;提高脑组织中SOD、GSH、CAT活性,降低MDA含量。结论拉莫三嗪对戊四唑大鼠点燃模型具有抗癫痫作用,并能提高点燃模型大鼠学习记忆能力及抗氧化能力。Objective: To study the effect of lamotrigine on the pentylentrazol (PTZ)-induced epilepsy model of rats. Methods: PTZ solution was injected in the rats to establish epilepsy rat models. When the rats had taken lamotrigine by peritoneal injection twenty eight days, the behavior and anti-oxidative condition were observed. From the twenty eight days to thirty one days, the behavior change in Morris water maze was observed. We measured the contents of SOD, GSH, CAT and MDA in the brain in the thirty two days. Results : The groups of lamotrigine prolonged delitesenee, reduced paroxysmal grade and durative time, decreased escape latency, improved the content of SOD, GSH and CAT and debased the MDA content at the same time. Conclusion: Lamotrigine has anti-epileptic seizure resistant function on PTZ-induced epilepsymodel of rats, which can elevate learning and memory and antioxidative capacity in rats.
