摘　　要：目的:建立小鼠淋巴瘤的原位脑肿瘤模型。方法:鼠源性B淋巴瘤细胞株A20,调整浓度到1×108 mL-1。在立体定向仪下将A20淋巴瘤细胞0.5μL植入BALB/c小鼠大脑中,每日观察,隔2d称体质量。3周后处死,制作石蜡切片做免疫组化及病理分析。结果:免疫组化及病理显示成功种植淋巴瘤,种植后小鼠体质量变化情况及存活时间恒定有规律。该方法建立的脑淋巴瘤动物模型在组织病理学上接近人脑淋巴瘤,且颅内生长稳定,成功率为80%。未见颅外转移病灶,实验周期短,重复性较好。结论:植入小鼠同源的B系淋巴瘤细胞株A20,可在小鼠大脑中形成原位肿瘤,其形态及恶性程度符合脑淋巴瘤模型的要求,生长特性及病理特征与人脑淋巴瘤相似,可作为临床淋巴瘤基础研究的理想模型。OBJECTIVE： To establish mouse primary central nervous lymphoma model. METHODS： The A20 cells and mouse B lymphoma cells line, were cultured in vitro and adjusted to the concentration of 1 ×10^8 cells/mL. The A20 cells solution as small as 0.5 uL was implanted into cerebral of BALB/c mice with the stereotactic apparatus. The living and survival states of mice with lymphoma were observed every day and weights were measured every two days. After 3 weeks of implantation, mice were killed and brain tumor was confirmed with common pathologic check and staining of CD20, Bcl-2 immunohistochemistry check. RESULTS： BALB/c A20 brain lymphoma models were confirmed by pathological checking and resemble histopathological features and immunohistochemistry feature of human brain lymphoma. The tumor loaded mice had regular change of weight and similar survival time. This kind of model was a reliant and reproducible, with 80% success rate as well as no extracranial growth extension. This kind of brain lymphoma of mouse had the advantage of short period of experiment. CONCLUSIONS： A20 lymphoma cell line can be implanted and develop to in situ tumor in BALB/c mouse＇s brain. This lymphoma model resembles histopathological and development features of human cerebral lymphoma, which can be used as a model to study the etiology and treatment of primary central nervous system lymphoma.
