原发性下肢静脉曲张与Tie2基因突变的关系

摘　　要：[目的]探讨原发性下肢静脉曲张（primary lower extremity varicose veins,PVV）与Tie2基因突变的关系.[方法]静脉曲张组为40例无家族遗传性（散发性）PVV患者,术中取剥脱的曲张静脉;正常对照组为18例行各类自体静脉移植手术的患者,术中取同部位正常静脉.用PCR方法对两组静脉标本第9号染色体Tie2基因的第14至23号外显子进行扩增,PCR产物直接送测序,测序结果同基因库中序列对比.[结果]正常对照组标本测序结果与基因库中Tie2基因序列一致,40例静脉曲张标本中均未发现Tie2编码区存在突变.[Objective] To explore the relationship between primary lower extremity varicose veins（PVV）and Tie2 gene mutation. [Methods] Varicose veins group was 40 sporadic PVV patients whose exfoliated varicose veins were taken in operation. Normal control group was 18 patients undergoing kinds of autogenous vein graft, and their normal veins were taken in operation. Tie2 gene of these samples were amplified by PCR and the 10 exons（number 14 to number 23） of Tie2 had been sequenced. The results were compared with gene bank. [Results] The sequence result of normal control group was the same as the sequence of Tie2 gene in gene bank. No mutation in the code region of Tie2 in 40 sporadic PVV patients. [Conclusion] Sporadic PVV may not be related with Tie2 gene mutation.
